Objective:To explore the relationship between the ERp44in placenta tissues and the pathogenesis of gestational diabetes mellitus.Methods:Sixty gestational diabetes mellitus patients (GDMgroup), hospitalized in Department of Obstetrics first affiliated hospital of Tai Yuan Wemen and children helth cenern from January2013to December2013, were selected as the subject group and60normal pregnant women as control group. Electron Microscopy was used to measure morphology of the endoplasmic reticulum in these women placenta trophocyte.(2)ERp44protein levels and cellular localization were measured by immunohistochemistry and Western blot, respectively, in term pregnancy placenta tissue.Results:(1) The ER within the syncytiotrophoblast was observed to be markedly dilated in GDM group compared with control group;(2)ERp44immunostaining in placenta tissue in GDM was positive, and slightly positive in placenta tissue in control group, statistically significant (p0.05).(3) Western blot, according to the results of two groups of placental villus tissues have two ERp44expression. Gestational diabetes group of grey value16.21±0.274,4.58±0.08in the control group, expression significantly increased, statistically significant (p<0.05).Conclusions:In women with gestational diabetes trophoblastic endoplasmic reticulumexpansion and swelling and obvious change; in the placenta of ERp44protein expression levels were significantly higher than in normal pregnant women. Suggest that endoplasmic reticulum stressassociated protein ERp44may be related with the onset of gestational diabetes mellitus.
