Cervical cancer is one of the most common malignant tumors among women overthe world, with about529,800new cases and275,100deaths each year.In developingcountries,453300new cases of cervical cancer and242000deaths were estimated everyyear,accounting for more than85%which in global.Generally the ends to the cervicalintraepithelial neoplasia(CIN)are:to normal or to cervical carcinoma. Trop-2,encodedby the positioning in1P32on TACSTD2(tumor-associated calcium signal transductiongene2),is a single transmembrane glycoprotein composed of323amino acids, and is acalcium channel signal converter, but its signal transduction specific mechanismmediated remains unclear.Currently the clinical study about relationships betweenTrop-2and cervical lesions has not been reported.In this study,the Trop-2expressionwas detected by RT-PCR and immunohistochemistry in cervical lesions and itsrelationship with clinical pathology was Investigated.Objective:To probe significance of Trop-2in the development of cervical cancerizationby observing the expression of Trop-2in cervical carcinoma, cervical intraepithelialneoplasia and normal cervix.Methods: Reverse transcription-polymerase chainreaction (RT-PCR) and Immunohistochemistry(IHC) were used to observe theexpression of Trop-2in normal cervix as control group, CIN Ⅰ and CIN Ⅱ, CIN Ⅲ,cervical carcinoma as experimental groups.The expression of Trop-2protein weredetected by IHC in respectively20cases and The expression of Trop-2mRNA wascharacterized by RT-PCR in respectively6cases. Results:RT-PCR showed aexpression of Trop-2in respectively group and the severer the disease was,the highermRNA expressed.IHC showed no expression of Trop-2protein in experimental group,whereas,in control groups,the expression of Trop-2is higher andhigher(p<0.01),which was located on the cell membrane.Conclusion:The expression ofTrop-2is linked to the occurred and developed of cervix cancer.In addition to be aclinical prognostic marker,Trop-2also may be a potential therapeutic target for cervixcancer.
