Objective: To investigate the significance of expres-sion of DNApolymerase delta catalytic subunit gene (POLD1) and its protein productp125in breast carcinoma.Method:1, collected69cases of clinically diagnosed breast cancer patientsafter surgical resection pathology of breast cancer and corresponding adjacenttissues;2, extract next69pairs of breast cancer and cancer tissue RNA, reversetranscription into cDNA as a template for fluorescence quantitative PCR, usingSYBR Green dye Ct value comparison method to detect the expression of breastcancer and adjacent tissues POLD1gene results with7500Software V2.0.5analyze data;3, with the Western Blot (Western blot) assay and the next69pairs of breastcancer tissue expression of p125protein in total protein using LI-COR Odysseyinfrared fluorescence imaging system scan scans, Odyssey v3.0software toanalyze data;4,Analysis of the correlation between the clinical data.Result:1, detected by fluorescence quantitative, gene expression levels inbreast cancer POLD1RQ=3.9±7.32, adjacent tissues expression was RQ=1.0 ±0.00. Detected by western blot, expression levels in breast cancer tissues was36.47±5.03; adjacent tissue expression was6.44±1.87. POLD1p125geneexpression in breast cancer and its encoded protein were higher than the adjacenttissues (p <0.05).2, further POLD1/p125and experimental analysis of breastcancer cases found in clinical TNM staging, POLD1/p125in the expressionlevels of0~Ⅱ period is higher in stage Ⅲ, Ⅳ(P <0.05). Both expressionlevels were diminishing with TNM.3, analyze the relationship between genesand clinicopathological features POLD1found POLD1genes and ki67positivecorrelation (P=0.007, p <0.05), while the ER, PR, Her2no significantcorrelation (P>0.05).Conclusion: Breast cancer associated with expression POLD1may also berelated to the malignant phenotype of the cell. Encoded p125protein wasdetected in favor Analyzing malignant proliferation of breast cancer cells isindicative of malignant lesions.
