Objective: Effects of p-CREB, CaMKII expression in manganesepoisoning mice and the protective effect of polygala tenuifolia willd inmanganese poisoning mice. Provide a theoretical and experimental basis for theprevention and treatment of manganese poisoning.Methods:Healthy adult60female Kunming mice were divided into5groups, the control group (CG), the manganese poisoning group (MG), themanganese poisoning and treated with gastric perfusion of high, medium, lowdosage polygala tenuifolia willd groups (MHG, MMG and MLG). The model ofmanganese exposed mice was manufactured by way of intraperitoneal injectionof manganese chloride.The ability of learning and memory was tested by Morriswater maze.The expression of p-CREB in CA1,CA3,DG of hippocampus andCaMKII in DG in mice was detected by immunohistochemistry. Analyze theaverage optical density of CaMKII and p-CREB by the image analysis system ofImage-Pro Plus6.0. To analyze the above data the method of one-way ANOVAResults:1. In place navigation test, compared with CG, escapetheincubation period in MG was significantly prolonged, the difference was statistically significant (P<0.01),the MHG, MMG, MLG groups comparedwith MG, the average escape latency decreased, the difference was statisticallysignificant (P<0.01), and compared with CG, the average escape latency change,there was no statistically significant difference (P>0.05).2. In spatial probe test,MG compared with CG group, timesofcrossing platform was decrease, thedifference was statistically significant (P<0.01), the MHG, MMG, MLGcompared with MG, the crossing platform significantly increased, the differencewas statistically significant (P<0.01), while compared with CG, times ofcrossing platform was no statistically significant difference (P>0.05).3. Inhippocampusarea CA1, CA3and DG: Compared with the MG, p-CREBprotein expression in hippocampus in MHG, MMG was significantly increased,the difference was statistically significant (P<0.01), while compared with CG,there was no statistically significant difference (P>0.05), the MLG comparedwith CG, the protein positive expression was to reduce, the difference wasstatistically significant (P<0.05).4. In hippocampal area DG the CG、MHG、MMG and MLG of hippocampus after CaMKII immunohistochemical positivereaction product of the average optical density value high than the MG, thedifference was statistically significant (P<0.01).5. Linear correlation analysisshowed that p-CREB expression in mice with the memory ability and theCaMKII expression of DG was positively correlated.Conclusions:1. The p-CREB and CaMKII.expression level inhippocampus of manganese exposed mice was higher by polygala tenuifoliawilld.2. The ability of learning&memory of manganese exposed mice wasimproved notably thepolygala tenuifolia willd.3. The p-CREB and CaMKIIexpression level in hippocampus of manganese exposed mice was low.4. That mechanism of the polygala tenuifolia willd can improve learning&memoryability in the manganese exposed mice might be associated with the increase ofexpression level of p-CREB and CaMKII.5. That mechanism of the polygalatenuifolia willd can improve learning&memory ability in the manganeseexposed mice might be associated with Polygala raised p-CREB upstreampassage CaMKII related.
