| Objective: Research the effect of oldenlandia diffusa injection on the Baxgene expression and proliferation of human osteosarcoma MG-63cells.Methods: Culture the human osteosarcoma MG-63cells in vitro, and testthe inhibitory rates of MG-63cells after24h with different concentrations ofoldenlandia diffusa injection by four methyl thiazolyl tetrazolium (MTT), thenobserve the morphology of cells by inverted microscope after the effect ofdrug. According to the results of the inhibition rates above, use the oldenlandiadiffusa injection with100ul/ml concentration to effect for6h,12h,24h, and48hrespectively, and detect the expression of Bax gene for each drug interventiongroup cells by RT-PCRResults: Oldenlandia diffusa injection can inhibit the proliferation ofMG-63cells, and prolonged with the increase in drug concentration and actiontime, which shows timeliness and dose effect. The MTT test displays the effectsof oldenlandia diffusa injection on the cells after24h with concentrations of12.5ul/mlã€25ul/mlã€50ul/mlã€100ul/mlã€200ul/mlã€300ul/mlã€400ul/ml: whenthe concentration is greater than50ul/ml, the inhibitory rate of MG-63cells are15.42±0.43%ã€67.21±0.27%ã€95.38±0.31%ã€98.73±0.56%and98.46±0.43%, and compared with the negative control group (P <0.05); when the drug concentration is less than25ul/ml, compared with the negative controlgroup, the inhibition rate shows no significant difference (P>0.05). Theinhibition rates of100ul/ml drug concentration for6hã€12hã€24hã€48h are:6.74±1.97%ã€44.38±2.31%ã€61.29±1.88%ã€87.53±2.13%, compared with thenegative control group (P <0.05). Under the observation of inverted microscope,the gaps among cells are larger while the density decreased and the cells appearto be morphological shrinkage, decrease the adherent ability, and the dead cellsbecame round floating in the culture medium.The results of detection on expression of Bax gene within the cells byRT-PCR show: compared with the control group, when the concentration is100ul/ml and with the prolongation of time, the Bax gene expression issignificantly increased (P <0.05).Conclusions: Oldenlandia diffusa injection can effectively inhibit theproliferation of human osteosarcoma MG-63cells, and the tumor suppressorfunction may be achieved by increasing the expression of tumor suppressorgene Bax. |
