Objective To study on low Period1expression in hepatocelular carcin-oma(HCC) and its correlation with poor prognosis of HCC patients afteroperation.Methods mRNA and protein expression of Period1were detected byreverse transcription polymerase chain reactionand western blot in30sampleswith HCC tissue and corresponding paracarcinomatous tissues.Period1promotermethylation level was detected by Methylation Specific PCR.Expression andlocalization of Period1protein were detected by immunohistochemistry in67paraffinembedded HCC samplesResults Compared to corresponding noncancerous tissue, cancer tissuehad decreased Periodl mRNA, protein levels and increased promotermethylation level in30samples (all P<0.05).The Period1positive expressionratio in67paraffin-embedded HCC samples was47.8%. Downregulatedexpression of Period1was related with multi-nodules,poor edmondson-steinergrade and vascular invasion.Conclusion Downregulated expression of Period1is closely relatedtumor development and metastasis in HCC. Promoter methylation is a important mechanism for downregulated expression of Period1.The loss of Period1expression may be a new tumor marker of poor prognosis in HCC.
