Objective: To establish of acute renal tubular injury model in rats byintra-peritoneal injection of gentamycin in the short period,so that then study theintervention of the drugs on the acute renal tubular injury provided modelsupport.Methods: Thirty Wistar rats of both sexes were randomly divided intothree groups: control group(n=10), GM1group(n=10), GM2group(n=10). Control group rats were intra-peritoneal injection of saline, for2days.GM1group rats were intra-peritoneal injection of gentamicin200mg/kg for2days.GM2group rats were intra-peritoneal injection of gentamicin400mg/kg for2days.24h urine volume, serum creatinine (SCr), urineN-acetyl-β-D-gluosaminidase (NAG) were determined at the end of experiment,and taked the nephridial tissue to observe renal pathological changes.Result: The24h urine volume,SCr and NAG level in GM2group wasmuch higher than that in control group and GM1group(Р﹤0.05).NAG level inGM1group was higher than its in control group(Р﹤0.05),24h urine volume and SCr level in that two groups with no significant differences (Р﹥0.05). Thepathological grading of injured renal tubule in GM1group were lower thanGM2group(Р﹤0.05).Conclusion:(1)Gentamicin can cause acute proximal renal tubules injuryin rats.(2)The400mg/kg/d of gentamicin dose were more easily to establish ofthe stable model of acute renal tubular damage in rats. Objective:To investigate the effect of prostaglandin E1ongentamicin-induced nephrotoxiciy in rats.Methods:Thirty Wistar rats of both sexes were randomly divided into threegroups:experimental group(n=10),contral group(n=10),blank group(n=10).20rats were intra-peritoneal injection of getamicin to establish model of acutetubular necrosis modol, prostaglandin E1or normal saline was administered for14days.10rats of blank group injected normal saline. Weight,24h urinevolume,serum creatinine(SCr), urine N-acetyl-β-D-gluosaminidase (NAG) weredetermined at day1,4,7,and14.The rats were sacrificed14days later,and takedthe nephridial tissue to observe renal pathological changes.Result:At day1,24h urine volume,SCr and NAG level in exper-imentalgroup and contral group was much higher than that in blank group(Р﹤ 0.05),with no significant differences between the former two groups(Р﹥0.05).At day7,SCr and NAG level after rising to the peak gradullydecile.Lower SCr and NAG were presented in experimental group comparedwith the contral group rats at day7and14(Р﹤0.05). The pathological gradingof injured renal tubule were lower than those contral group(Р﹤0.05).Conclusion:(1)PGE1can reduse SCr and NAG level in rats thatgentamincin induesd nephrotoxicity,alleviate function of rennal.(2)PGE1cnalleviate renal pathological tissue damaged and promote renal tubular epithelialcells regeneration.PGE1and provide a certain degree of protection for renal.
