Tongue squamous cell carcinoma, which are the most common malignant tumors of oral cancers, display a high degree of epithelial differentiation potential. Generally, Cancer cells tend to infiltrate into the surrounding normal tissue, such as neural infiltration, intravascular tumor, as well as metastasis of cervical lymph node which often appear on the same side. However, when the lesion is located in or involved to the midline, then there is a metastasizing tendency to contralateral or bilateral regions. To determine the optimal treatment, the options are based on the TNM stage of tumor size, location and metastatic spreading range. At present, the main therapeutic schedule is comprehensive and sequential therapy which combined surgery with radiotherapy and chemotherapy. Despite all this, many scholars has diverted attention to personalized targeted gene therapy because of the poor prognosis and functional recovery.The objective of this study was to research the influence of lentivirus-mediated cyclooxygenase-2gene by RNAi technology on the proliferation and invasion of Tca8113and to investigate the mechanism of lentivirus-mediated transfection of COX-2gene on tongue squamous cell carcinoma.The frist step was to select the stable living Tca8113cell lines and to set up COX-2gene silencing group (pLKO.1shCOX-2), non-carrier negative group (Tca8113scr) and Tca8113cell group. And then conventional cell culture on three groups of cells.The western-blot was used to verify the productiveness of silence on COX-2protein and the cloning formation assay was used to examine the transformation of tumorigenicity.The silence of cyclooxygenase-2gene on invasion and metastasis changes were detected by Transwell chamber of the invasion and migration experiments.Collectively, these data demonstrated the conclusions as follow:1. The western blot data showed that a significantly reduced in Tca8113transfected by lentivirus-mediated. 2. The silent COX-2treated would effectively inhibit the tumorigenicity Tca8113.3. The ability of invasion and metastasis can be suspessed after lentivirus-mediated COX-2gene silence.
