| Objective:To detect the expression of MIF protein and mRNA in the Hepatocellular carcinoma(HCC), hepatitis B liver cirrhosis and normal liver tissues. While tested the expression of pERK and pJNK in the MAPK signaling pathway. Preliminary discussion the possible mechanism of MIF facilitate hepatitis b cirrhosis into HCC through MAPK signaling pathways.Methods:Fifty-two HCC specimens and corresponding adjacent cirrhosis tissues as experimental group, twenty normal liver tissues as control group.All tissue were derived from department of hepatobiliary surgical in Lanzhou general hospital, Lanzhou Command. All of patients in the experimental group were not receive radiotherapy, chemotherapy or any other anti-tumor therapy. All tissues samples were fixed with10%formaldehyde, embedded within paraffinand, made into tissue microarray, and then made into4μm thick slices. Using immunohistochemistry to detect the expression level of MIF, ERK1,JNK1protein in52cases of HCC tissue, para-cancer cirrhosis tissue and twenty cases of normal liver tissues,and the expression level of pERK, pJNK protein.. The expression level of MIF mRNA and ERK1mRNA and JNK1mRNA by In situ hybridization(ISH) in all tissues.Results:The positive expression rate of MIF protein showed by immunohistochemistry in HCC, cirrhosis and normal liver tissues was78.85%,75.0%,20.0%respectively. The positive expression rate of ERK1protein showed by immunohistochemistry in HCC, cirrhosis and normal liver tissues was80.77%,71.75%,35.0%respectively. And the positive expression rate of JNKI protein showed by immunohistochemistry in HCC, cirrhosis and normal liver tissues was73.08%,67.31%,25.0%respectively. The positive expression rate of pERK protein showed by immunofluorescence in HCC, cirrhosis and normal liver tissues was75%,46.15%,20.0%respectively. The positive expression rate of pJNK protein showed by immunofluorescence in HCC, cirrhosis and normal liver tissues was67.31%,44.23%,20.0%respectively.The difference had statistically significance of MIF, ERK1, JNK1protein expression among HCC, HBV liver cirrhosis compared with normal liver tissues(P<0.05), there is no difference between HCC and HBV liver cirrhosis (P>0.05). While,there are statistically differences of pERK,pJNK protein expressions among HCC compared with HBV liver cirrhosis and normal liver tissues(P<0.05), there is no difference between HBV liver cirrhosis and normal liver tissues (P>0.05). ISH showed that the positive expression rate of MIFmRNA in HCC, liver cirrhosis and normal liver tissue was69.23%,71.75%,20.0%respectively. The positive expression rate of ERK1mRNA in HCC, liver cirrhosis and normal liver tissue was76.92%,78.85%,35.0%respectively. And that the positive expression rate of JNK1mRNA in HCC, liver cirrhosis and normal liver tissue was75.0%,71.75%,30.0%respectively. The difference had statistically significance of MIFmRNA, ERK1mRNA, JNK1mRNA expression among different liver tissues(P<0.05).Conclusion:The overexpression of MIF, ERK1, JNK1are associated with the development of hepatitis B liver cirrhosis. And closely correlated with the occurrence of HCC, it maybe through the MAPK signaling pathways mediating hepatitis B liver cirrhosis into HCC, there need for further analysis and research. |
