Leakage is the most important reason for the failure of root canal treatment.Inthis study, we compared five different root canal sellers which were commonly usedin clinic.There were zinc oxide-containing sealer,resin sealers and silicone-basedsealer,and so far, there were no studies to compare their sealing abilities.Objective To compare the apical sealing ability of five root canal sealers-EZFill,AH Plus,2Seal,Cortisomol and GuttaFlow by using a glucose leakage test.Methods62extracted mandibular premolars with single root canals wereprepared with Protaper instruments to F3. After each instrument, the canals wereirrigated with5.25%sodium hypochlorite, and after the completion of the root canalpreparation,the canals were irrigated with17%EDTA and5.25%sodium hypochloriteto remove smear layer.Then the teeth were randomly divided into five groupscontaining10teeth each. Additionally, the negative and positive control groupcontained6teeth in each group. Ten root canals were each filled with EZ Fill,AHPlus and2Seal sealers with warm vertical obturation technique to finish root canalfillings;16root canals were filled with Cortisomol sealers with warm verticalobturation technique to finish root canal fillings(6root canals used as negativecontrol group);and the in GuttaFlow group,GuttaFlow is triturated in its cannulaand passively injected into the canal and then used with single gutta-perchapoints.Then,2layers of nail varnish was applied to all external tooth surfaces exceptfor the coronal access cavity and the apical end of the root in the control andexperimental groups.The entire specimen was coated with2layers of nail varnish,including the the apical foramina.Subsequently,a modified glucose leakage modelwas made and the amount of leakage was determined after1,2,4,6and8weeks byglucose oxidase colorimetric method.Results Negative controls revealed no fluid movement and positive controlsrevealed high leakage.At1week,compared with AH Plus,Cortisomol showed higherleakage(P<0.05),and at2and4week,compared with EZ Fill,AH Plus,2Seal andGuttaFlow, Cortisomol showed higher leakage(P<0.001),and the leakage was not significantly different among EZ Fil,AH Plus,2Seal and GuttaFlow(P>0.05).But at34and48days, compared with EZ Fill,2Seal,Cortisomol andGuttaFlow, AH Plus showed lower leakage(P<0.05),and the differences betweenEZ Fill,2Seal,Cortisomol and GuttaFlow was not significant(P>0.05).Conclusion In the early after the obturation of root canal, Cortisomolshowed more leakage than EZ Fill,AH Plus,2Seal and GuttaFlow. In the later, thesealing abilities of EZ Fill,2Seal Cortisomol and GuttaFlow showed no significantdifferences,but less leakage was detected with AH Plus than Cortisomol、EZ Fill、2Seal and GuttaFlow. So in the clinical,considering the effect of leakage to thelong-term outcome of root canal treatment, clinicians had better choose resin sealers,especially AH Plus.
