Objects:The aim of the study was to evaluate the expression levels of CRM1and Her-2/neu in gastric cancers and the corresponding normal tissues, correlation with their clinicopathological parameters and the relationship of CRM1and Her-2/neu in the metastasis and occurrence gastric carcinoma.Methods:135postoperative patients were examined for CRM1expression by immunohistochemistry and for Her-2/neu expression by fluorescence in situ hybridization (FISH).Results:The positive expression rate of CRM1in neoplastic tissues and adjacent gastric mucosa were57.8%and6.7%respectively. CRM1levels were linked to more advanced tumor stages, poor prognosis, and likelihood of recurrence (P<0.05). The Cox multivariate analysis indicated that the CRM1expression was an independent indicator for tumor recurrence (P=0.0048).16of the135gastric cancer specimens (11.85%) were positive for Her-2/neu, the expressions were not found in paired normal tumor-adjacent. Positive cases for Her-2/neu also had high CRM1expression levels (P=0.0104).Conclusion:The current results suggest that CRM1is over-expressed in gastric carcinomas and is related to poor patient outcome. HER-2/neu gene has a higher amplification proportion in gastric, and has a certain correlation with the CRM1expression:HER-2/neu patients with high expression of CRM1was amplified. Therefore, the relationship of CRM1and Her-2/neu expression in gastric cancer may help us evaluate the biological behavior of gastric cancer correctly and be benefit for selection therapeutic approaches.
