Objective:Through the production of new medical biofilm materials, to research making methods, and to evaluate its biocompatibility., then get ready for clinical application.Methods:Under sterile conditions, take out the randomly selected healthy porcine pericardium, decellularize cells via two methods, which are descaling with trypsin, repeated freezing and thawing with nucleic acid enzymes,to prepare pig-pericardium medical membrane. We detect excellent pig-pericardium medical membranes through microstructure, then evaluate its biocompatibility and absorption time via rat subcutaneous embedding methods experiments.Results:(1)No distinctly difference is between the two pig pericardium medical biofilm and fresh pig pericardium through in naked eyes. All biofilms are complete and smooth.while the two type which acellular ones are pale.(2) Under the light microscope,the constitute of pig pericardium medical biofilm is visible clearly. Some cells residues can be seen in some areas of serous layer, most loss. There is arranged in bundles in various directions of collagen fibers in fibrous coat, which collagen fiber bundle is ranked no apparent disorder, wave-shaped, without dissolving and breaking. Cells are almost completely removed by trypsin method, and repeated freezing and thawing with nucleic acid enzymes method, while controlling group has a mount of cells left.(3) No antibiotics was used on the rats after operation. No rat was infected. Wound healed naturally, without redness and discharge of foreign bodies. Rat has not local, systemic and behavior abnormal. No rat was dying or dead.(4) There is no significant change in microstructure of the pig-pericardium in a-week. Two weeks later fiber started breakage.Four weeks later, lymphocytes began to invade. Leukocyte infiltrate largely in specimens after eight weeks.Conclusions:(1)The pig-pericardium medical biofilm which is made by trypsin is more aviliable as it meet our requirements.(2) Pig-pericardium biological material is much more ideal alternative materials membrane,which is making easy, inexpensive, and owns good biocompatibility.
