Objective to discussion if Amifostine(AMI) has the protective effect for tumor after radiotherapy or if it has influence to radiotherapyMethods to culture the Lewis Lung Cancer Cells, when the cells cover80%of the culture apparatus, subculture the cells, then get cell suspension with106/L. Intramuscular inject0.2mL cell suspension on the right lower limbs of the mice, after2-3days, the tumor come into formation and the tumor model are preparation. divide the mice into four groups:AMI and RT(A group), RT(B group),AMI(C group), Contrast(D group),make sure there are8mice in every group.Before radiotherapy,give group A and group C AMI200mg/Kg intraperitoneal injection, the same dose normal saline for group B and D, intraperitoneal injection.30minutes later give group A and group B radiotherapy (6M-X,10Gy). Measuring the Change of mice weight and tumor volume every two days. Fifteen days later kill the mice, got the apoptosis of tumor cells with Tunel.Results There are not significant differences among every group on the mice weight (p>0.05).At the frist day, the tumor volume among every group are not significant differences(F=1.201, p>0.05).From the third day to the thirteen day the tumor volume in group A are less than the other3groups, and the tumor volume in group B are less than group C and group D, there are significant differences (F=15.47, q=0.2809-9.6726,p<0.05). There are not significant differences on the tumor volume between group A and group B, also between group C and group D. The cell apoptosis in group A are higher than that in the other3groups, the cell apoptosis in group B and group C are higher than that in group D, There are significant differences(F=36.535, q=0.6875-8.3816, p<0.001), between group B and group C the cell apoptosis are not significant differences.Conclusion Amifostine protects normal cells from radiation damage without extenuation of tumoricidal potential against Lewis lung cancer, furthermore Amifostine might enhance the apoptosis of tumor.
