Objective:To research the effect of cannabinoid receptor2in lung injury,liver injury and kidney injury induced by severe acute pancreatitis in rats.Method:Ninety-six SD rats (SPF) were divided into four groups randomly. Sham group: The pancreatic tissue of rats were fliped after laparotomy and then closed the peritoneal cavity. SAP group:4%sodium taurocholate was injected into pancreaticobiliary duct to establish SAP model. HU-308group:1hour later after the SAP model established, HU-308was injected into peritoneal cavity by2.5mg/kg. AM-630group:1hour later after the SAP model established, AM-630was injected into peritoneal cavity by2.5mg/kg. At3,6,12h,4to5ml blood was drawed from carotid artery and then centrifuged to get the serum,the lever of AMY, TNF-a, IL-6, AST, ALT, BUN and Cr was measured in it. The left lower lobe of lung tissue was grinded to slurry, the MPO was measured in it, the right lower lobe of lung tissue was made to pathological sections and scored. The last lung tissue weighted, then baked in the oven for72hours, the dry tissue was weighted.Result:At each time, compared with Sham group, the leves of AMY, TNF-a, IL-6, MPO, pathological score and wet to dry ratio in HU-308group, AM-630group and SAP group were higher (P<0.05), At3h, compared with the Sham group, the leves of ALT, AST, BUN, Cr in each of three groups have no statistically significant difference (P>0.05), at6h and12h, the leves of ALT, AST, BUN, Cr in three groups were higher than the Sham groups’(P<0.05). At3h, the leves of AMY, TNF-a, IL-6, MPO, pathological score and wet to dry ratio in HU-308group were lower than the SAP groups’(P<0.05), the leves of ALT, AST, BUN, Cr were no statistically significant difference with the SAP groups’(P>0.05), at6h, compared with SAP group, the leves of AMY, TNF-a, IL-6, MPO, pathological score.wet to dry ratio ALT, AST, BUN, Cr were lower (P<0.05). At3h, the leves of AMY, TNF-a, IL-6MPO were higher than the SAP groups’(P<0.05), the leves of wet to dry ratio, pathological score, ALT,AST, BUN, Cr were no statistically significant difference with the SAP groups’(P>0.05), at6h, the leves of AMY, TNF-a, IL-6, MPO, wet to dry weight ratio, pathology score, AST, BUN and Cr were higher than the SAP groups’(P<0.05), the leve of ALT was no statistically significant difference with the SAP groups’(P>0.05),at12h, the leve of each one were higher than the SAP groups’(P< 0.05).Conclusion:The CB2receptor whice was activatied played a protective role in lung injury,liver injury and kidney injury induced by severe acute pancreatitis. On the contrary, the CB2receptor inhibited increase the degree of injury.
