Experimental Study Of Haikunshenxi Capsule And Bliling Capsule On Hyperuricemic Nephropathy

Objective:To study the effect and possible mechanisms of Haikunshenxi capsule and Bailing Capsule on hyperuricemic nephropathy.Methods:90male Wistar rats were randomly divided into four groups, including normal group, hyperuricemia group, haikunshenxi group and bailing group after a adaptive feed feeding week. The rats in hyperuricemia group were fed with10%yeast diet, and administered intragastrically with adenine (50mg/kg.d), subcutaneously giving oxygen oxazine acid potassium suspension4ml/(kg.d) at the same time. Intervention group at the same time give corresponding drug lavage therapy. After2,4and6weeks, six rats was randomly selected from each group. We collected the angular vein blood andurine and24h urine for detection of uric acid (UA), urea nitrogen (BUN), creatinine (CR) and creatinine clearance rate (Ccr) and urinary microalbuminuria. Two rats were ecuted in each group for renal tissue HE and MASSON staining and immunohistochemical determination of interstitial fibrosis(IL-1β), transforming growth factor (TGF-β1), and a-smooth muscle actin(a-SMA) expression,and counting left kidney mass index.Results:Compared with normal group, hyperuricemia group after2,4,6weeks UA, BUN, CR,24hours urinary protein, left kidney mass fraction increases, the Ccr is reduced, the result was statistically significant (p<0.01). Compared with hyperuricemia group, haikunshenxi group after2,4,6weeks CR, Ccr,24hours urinary microalbuminuria increased (p<0.01), after6weeks BUNand left kidney mass index have obvious difference (p<0.01); bailing group after2,4,6weeks Ccr,24hours urinary microalbuminuria have obvious increased, after2and6weeks BUN have obvious difference, afer4and6weeks CR and left kidney mass index have obvious difference (p <0.01).Compared with the normal group, hyperuricemia group after2,4,6weeks HE and MASSON staining prompt increased renal interstitial collagen fibre, renal interstitial tissue expression of TGF-βb, IL-1β,α-SMA significantly increased (p<0.01). Compared with hyperuricemia group, haikunshenxi group showed siginificantly lower TGF-β1, IL-1β, a-SMA expression, along with renal interstitial fibrosis after2,4,6weeks; bailing group showed TGF-β1, IL-1β, a-SMA expression significantly decreased after4,6weeks(p<0.01).Conclusions:Haikunshenxi capsule and Bailing Capsule can partly improve rat renal function, and reduce TGF-β1, IL-1β, a-SMA expression in the kidney in rats.
