Objective To investigate the taxonomic richness, diversity and distribution changes of gut microbiota in sporadic colorectal cancer with adenoma patients.Methods We collection the the colorectal cancerous tissue, para-cancerous tissue, matched noncancerous normal tissue, adenoma tissue and stools of21CRC with adenoma patients who underwent different surgical procedures between January,2013and August,2013at the Affiliated Hospital of Qingdao University.454pyrosequencing was performed on a454Genome Sequencer FLX platform.Results Our investigations implicated that differences between individuals were significantly greater than intrasubject differences. In addition, the fecal microbiota had a significant difference from intestinal tissue. We observed a positive correlation between the abundance of genus Fusobacterium, Campylobacter, Howardella and the procedure of CRC disease; and a negative correlation of genus Arthrobacter; another positive correlation between the abundance of genus Fusobacterium, Campylobacter and the distance from the tumor, and a negative correlation of genus Arthrobacter; a positive correlation between the abundance of genus Faecalibacterium, Oscillibacter and the CRC disease status, and a negative correlation of genus Streptococcus.Conclusion The structural of gut microbiota changed significantly in CRC with adenoma patients that gradual increase of harmful opportunistic pathogens in normal, adenma, cancerous tissues or normal, paracancerous, cancerous tissues, while gradual reduction of probiotics bacteria. The abundance of certain key intestinal bacterial phylotypes gradual change may play an important role in the procedure of colorectal cancer.
