Investigation On Adaptation Culture Of Japanese Encephalitis Virus In Vero Cells And Pilot Scale Culture

To complete the adaptation and stability research of Japanese encephalitis virus(JEV) in Vero cells, and find the best culture conditions.The JEV P3strain for production of vaccine was cultured continuously for20passages in Vero cells for determination of virulence and for RT-PCR analysis. TheJEV P3strain had great adaptability in Vero cells.The condition for stationary culture of JEV, such as pH value of maintenancemedium, culture temperature, multiplicity of infection (MOI) and culture ways wereoptimized. At last, the condition for stationary culture of JEV, such as pH value ofmaintenance medium, culture temperature, multiplicity of infection (MOI) and cultureways were optimized.The master and working seed libraries of JEV were set up and tested by referenceChinese Pharmacopoeia2010edition. The master and working seed libraries of JEVwere set up successfully and the results of the checks were in line with the ChinesePharmacopoeia2010edition. The master and working seed libraries of JEV can beused for the research of inactivated Japanese encephalitis vaccine.
