Objective:Construction of methicillin-resistant staphylococcus aureus biofilm in vitro model,to explore san huang shao shang ling and radix scutellariae’s impact on the biofilm.Methods:Build MRSA biofilm in vitro model:automatic bacteria analyzer and disc diffusion method for MRSA,applicating new blue-Congo red stain qualitative MRSA biofilms,with polyvinyl chloride (PVC) chip as the carrier,96orifice MRSA biofilm in vitro model is set up.Using scanning electron microscope observation of biofilm formation, application of crystal violet biofilm semi-quantitative,respectively in the cultivation of1d,3d,5danalysis its ability to form biofilms,in3days of modeling (mature biofilm) were determined using orifice plate spirit extract radix scutellariae,the MIC value of the clinical MRSA. Drug effect after24hours, radix scutellariae, san huang shao shang ling observed by a scanning electron microscopy(sem)extract on mature MRSA resistant effect of biological membrane.Results:Ali new LAN results of Congo red staining and scanning electron microscopy (sem) and semi-quantitative positive rate was100%, the enzyme standard meter crystal violet semi-quantitative and scanning electron microscope showed MRSA biofilms in order to appear1d,3d and5d stability,three yellow burns spirits containing radix scutellariae (425)ug extract (2048)ug and radix scutellariae can damage to mature MRSA organisms,and san huang shao shang ling living bacterium number from radix scutellariae group decreased significantly.Conclusions:Ali new LAN-Congo red stain is rapid and accurate methods to curtain biofilm qualitative; compared with the single taste traditional Chinese medicine radix scutellariae, San huang shao shang ling can effectively destroy MRSA biofilms,which has obvious advantages,not only can more effectively destroy biofilms,and can kill bacteria better.
