ObjectiveTo study the expression of nuclear factor-κB (NF-κB), P-gp, CYP3A4in the tissueof diffuse large B-cell lymphoma (DLBCL), in order to explore their relationship withbiology behavior such as pathological staging, tissue typing in DLBCL. Combined withthe follow-up data, to analyze the relationship between the expression of NF-κB, P-gp,CYP3A4and prognosis in order to provide theoretical basis for gene targeting treatmentof DLBCL.MethodsThe expression of NF-κB/p65, P-gp, CYP3A4in the tissue of DLBCL andlymphoma hyperplasia were detected by using immunohistochemical staining, thecorrelation between three indicators above and biology behavior such as age, gender,pathological staging, tissue typing and extranodal involvement sites in DLBCL wereanalyzed. Correlation analysis was conducted to analyze the relationship betweenNF-κb/p65, P-gp, CYP3A4in DLBCL tissue. Kaplan-Meier was performed to analyzetherelation between the level of NF-κb/p65, P-gp, CYP3A4and prognosis of DLBCL.Results1. The positive expression rate of NF-κB/p65in DLBCL and lymphomahyperplasia was62.8%,23.3%, respectively, it was significantly different (χ2=13.89,P<0.05). The positive expression rate of NF-κb/p65increased as the increase of IPIscore, Ann Arbor stage increased (P<0.05), but nothing on age, gender, tissue typing,extranodal involvement sites and level of LDH (P>0.05). 2. The positive expression rate of P-gp in DLBCL and lymphoma hyperplasia was57.0%,20.0%, respectively, it was statistically significant (χ2=12.20, P<0.05). Thepositive expression rate of P-gp increased as the increase of extranodal involvementsites, Ann Arbor stage increased (P<0.05), but not related to age, gender, tissue typing,IPI score and level of LDH (P>0.05).3. The positive expression rate of CYP3A4in DLBCL and lymphoma hyperplasiawas33.7%,13.3%, respectively, it was statistically significant (χ2=4.54, P<0.05). Thepositive expression rate of P-gp increased as the increase of IPI score, Ann Arbor stageincreased (P<0.05), but not related to age, gender, tissue typing, extranodal involvementsites and level of LDH (P>0.05).4. The expression level of NF-κB/p65incarcinoma tissue in DLBCL was positivecorrelated with P-gp(r=0.593, P<0.05), and there was the same correlation betweenNF-κB/p65and CYP3A4(r=0.452, P<0.05), while there was no significant relationshipbetween P-gp and CYP3A4(r=0.155,P>0.05).5. The two-year survival rate in NF-κB/p65positive group was25.9%, which wassignificantly lower than that in NF-κB/p65negative group68.8%(P<0.05). Thetwo-year survival rate in P-gp positive group was36.7%, which was significantly lowerthan that in P-gp negative group64.9%(P<0.05). The two-year survival rate inCYP3A4positive group was31.0%, which was significantly lower than that inCYP3A4negative group68.4%(P<0.05).Conclusion1. The expression of NF-κB/p65, P-gp, CYP3A4in the tissue of DLBCL are higherthan those in lymphoma hyperplasia.2.The positive rate of NF-κB/p65, P-gp, CYP3A4was increased with the increaseof clinical stage.3. There was remarkably correlation among NF-κB/p65, P-gp, CYP3A4in thetissue of DLBCL, they are involved in the process of the development and metastasis ofDLBCL.4. The prognosis of NF-κ B/p65, P-gp, CYP3A4positive expression in DLBCLtissue is significantly worse than that of negative expression, suggesting that positive expressions of NF-κ B/p65, P-gp, CYP3A4are indexes for adverse prognosis.Detectionof the expression of these indicators has important reference value in the judge ofmalignant degree and prognosis of DLBCL.
