Tea not only has the characteristics of natural health, also has more health and nutrition affect. Tea contains protein, carbohydrates, lipids; and rich in tea polyphenol, caffine, tea polysaccharide, tea saponin, theaflavins, vitamins, trace elements and minerals etc. The study found that the natural functional components of tea have the effect, such as:antioxidant and UV protection, obesity, cardiovascular disease resistance, anti-inflammatory.anti-tumor.This experiment use tea polyphenols, EGCG, theanine and caffeine, theaflavins, tea pigment on human breast cancer MCF7cells in vitro. We find the most safe and effective-EGCG, for nano modification, preparation of nano particles. The preparation of the nanometer particles on MCF-7cell experiment again. Compare the distinguish of difference modified EGCG-NPs:FA-EGCG-NPS. PEG-EGCG-NPS (FA+PEG)-EGCG-CSNPS-1and (FA+PEG)-EGCG-NPS-2.The results are following:1, Using inverted microscope to observe the morphology of human breast cancer MCF-7cell which was treated with Polyphenols, EGCG, theanine, caffeine, theaflavins and tea pigment.The same dose, inhibition rate shows as the following:theaflavins>EGCG>caffeine>tea polyphenols> tea pigment> theanine.2, In order to improve the bioavailability of EGCG, we use nanotechnology to optimize EGCG, make up for its disadvantages and achieve dose advantage. Nano-EGCG were makered by Rhodamine B. We use laser confocal microscopy to observe the targeting property of the nanoparticles.Confocal images of MCF-7cells after incubated24h with EGCG nanoparticles. The best is FA-EGCG nanoparticles, and have folic acid modified nanoparticles targeting higher than undecorated particles.3, Using inverted fluorescence microscope nanoparticles shoot MCF-7cell morphology after incubated24h,48h,72h with EGCG nanoparticles.The best effect is (FA+PEG)-EGCG-NPs. Nanoparticles in the experimental concentration range (20,40,80,160,240,320,400μg/mL) have no cytotoxicity.4, Treated MCF-7cells with EGCG、(FA+PEG)-EGCG-CSNPS-1and (FA+PEG)-EGCG-NPS-2, after48h, extract total protein in cells.Western-Blotting shows, protein level of p-PDK1,p-Akt,CyclinDl, Bcl-2were down, and PTEN、p21、Bax were up regulated in MCF-7cells when treated by240μ g/mL EGCG,(FA+PEG)-EGCG-CSNPS-1and (FA+PEG)-EGCG-NPS-2.
