| Inonotus obliquus (Fr.) Pilat is a kind of precious medicinal fungi, which in our country are mainly distributed in the Greater Hinggan Mountains area of Heilongjiang and Jilin Province. In recent years, the scholars had further studied the active ingredient of Inonotus obliquus, to use for cancer, diabetes, cardiovascular disease, liver disease, and AIDS prevention and treatment. At present, the research of Inonotus obliquus in China has still been in its infancy. This topic was the first time for the Inonotus obliquus on mice acute toxicity experiments. The effect of extracts in Inonotus obliquus on diabetic mice was researched by oxidative stress, chronic inflammation and lipid metabolism, which effective role of specific components were further studied.The experimental type â…¡ diabetes model mice were replicated by STZ-induced method. The Metformin and Xiaokeling tablets were positive drug. The mice was administrated for4weeks. The average water intake, feed intake, the weight and FBG of mice were weight during the experiment.2hours after the last dosing, the blood was draw and centrifuged. The blood cell classification, TC, TG, HDL-C, LDL-C, were detected with automatic biochemical analyzer. The SOD, MDA, GSH-Px, GSP, FINS, TNF-a, IL-8and IL-10were detected, and the organ index was calculated. The water extract and alcohol extract in Inonotus obliquus could reduce the general symptoms of diabetic mice, FBG, FINS, GSP, TC, TG, WBC and PLT. Compared with alcohol extracts, the effect of water extracts in treating experimental type â…¡ diabetic mice was better.The Inonotus obliquus crude polysaccharide was extracted with water extraction-alcohol precipitation. The positive medicine was Xiaoke pills. Combined with whole animal behavioral experiments, optical mouse tail-flick test and physical evaluation, the comprehensive treatment of Inonotus obliquus polysaccharide on diabetes was studied. The Inonotus obliquus polysaccharide could reduce FBG, FINS, GSP, the number of spontaneous activity and the latency of tail-flick optical stimulation, improved load exhaustive swimming time.The results showed that:Inonotus obliquus extract could obviously sharpen the oxidation resistance ability and physical fitness of model mice, and regulating lipid metabolism, inhibiting the inflammatory response, easing peripheral nerve lesions. Compared with alcohol extracts, the effect of water extracts in treating experimental type II diabetic mice was better. The effect of active ingredient is not just polysaccharides, but also include other ingredients. |
