Objective:Based on the textual research of Sichuan Cirsium Mill.,this thesis has made a comparison of C. japonicum, C. Setosumand analogues on crude drug characters and anatomical characteristics, then did qualitative and quantitative analysis of linarin and chlorogenic acid as index component by TLC, HPLC. This thistle put aims on exploring the phylogenetic relationship of Sichuan medicinal Thistle plants and the correlation between active chemical components, to provide theoretical and experimental basis for quality control of C. japonicum and C Setosumand.Methods:Textual research and discuss of6Sichuan Cirsium Mill, species and Olgaea leucophylla, which is the analogue of C. japonicum, has done through method of literature materials consulting, and the taxonomic status of these7species have been confirmed by the means of morphological classification. Cluster analysis of the micro morphology data has done by the SPSS17. The root, stem and leaf cross-section characteristics of these7species give the medicinal materials identification of C japonicum and C. Setosumand more references. Qualitative and quantitative analysis of linarin as index component can make the identification of similar products more accuracy.Results:7species medicinal materials have been researched in this study,5of them are used as C. japonicum, but the C. Setosumand has no this mixing-use phenomenon. The tree diagram of micro morphological characteristics showed:C handelii and C. leducei were in one group, C. japonicum and C.leo in another group, but Olgaea leucophylla was entirely separate from the other species,this result is basically the same as the classical classification.TLC showed that there were bigger difference in spots number and the Rf of7medicines, the C. racemiforme and Olgaea leucophylla have no buddleoside. HPLC results show that the Olgaea leucophylla does not contain buddleoside, chlorogenic acid, and the other6materials contain chlorogenic acid, but the C. racemiforme has no buddleoside. pectolinarin was only detected in C. japonicum toot and C. leducei.Conclusion:Cirsium Mill, medicinal plants have been mixed-use and misused on a certain degree, and C. japonicum is also a polymorphic specie,which made the identification of the medicinal materials more difficult.The root, stems and leaves’cell anatomical features of C. japonicum and the similar plants can enrich the identification technical indicators for herbs, and also can be one way to distinguish the authenticity of herbs. Cluster analysis of micro morphological can give a phylogenetic relationship diagram which is basically the same as taxonomic status in Sichuan Cirsium Mill., which means the micro-morphological characteristics of herbs can be used as identification method for C. japonicum and C. Setosumand herbs. C. japonicum and C. Setosumand are similar in chemical composition, so the mixing-use in ancient have certain basis. But both differences in morphology and TLC graphics, are supportting modern science of Chinese materia medica to use respectively. Chlorogenic acid may be the corporate chemical composition of Sichuan medicinal Cirsium Mill, plants, and Olgaea leucophylla is Olgaea Iljin plant dose not contain chlorogenic acid for it is belong with Olgaea, C.leo, C. leducei and C. handelii are the similar products of C. japonicum, and they are often used confusedly, but TLC and HPLC showed that should make the medicine material. C. racemiforme and Olgaea leucophylla are quite different in chemical component,and they should be strictly distinct as fakes。...
