ObjectiveTo treat the injury of sciatic nerve combined with the nerve mobilization which with different stretching parameters.By testing triceps surae wet weight, and testing ubiquitin protein ligase (Murf-1) detection,in order to observe the impacts of denervation muscular atrophy which were treated by nerve mobilization,while obtaining the optimal stretching parameter.So we can provide a theoretical basis for clinical practice.Methods1.After6Japanese rabbits were anesthetized,completely exposed the sciatic nerve on the outer-mid thigh.Then marked a2cm segment of nerve with surgical sutures,setted restrictive factors,prepared the ready posture.Made a knee-extension activities slowly and passively which along the sciatic nerve route.When the marked segment was extended to the required length parameter,recorded the angle of knee.Obtained the mean after6rabbits repeated the process.These values were the standard interventions of nerve mobilization.2.To divide30rabbits into sham operation group,model group and nerve mobilization group randomly.According to the differernt stretching parameters,dividing the nerve mobilization group into three subgroups.Nerve mobilization group A had the stretching parameter of6%, nerve mobilization group B had the stretching parameter of9%, nerve mobilization group C had the stretching parameter of12%.Each group had6rabbits.Forceps was used to make the model of acute injury of sciatic nerve.3days after operation,we began to interve without sham operatin group and model group.After28days, wet weight testing of triceps surae, and Western Blot were used to clarify the influence of nerve mobilization to the prevention and treatment of denervation mucular atrophyResults1.Angles of nerve mobilization with different stretching parametersrthe nerve mobilization group with stretching parameter of6%,its angle was approximately90°; the nerve mobilization group with stretching parameter of9%,its angle was approximately130°; the nerve mobilization group with stretching parameter of6%,its angle was approximately170°.2.Wet weight of triceps surae:besides both sides of the sham operation group had no significant difference(P>0.05),there was a obvious difference in the both sides of the rest groups(P<0.05).Operation side:compared with the sham operation group,wet weight of the model group and all nerve mobilizaiton groups were obviously decreased,which with statistical meaning(P<0.05);compared with the model group,the wet weight of both group A and B were heavier than the model group,the differences were significant(P<0.05),while there were no statistically significances between the group C and the model group(P>0.05);the compare among three groups,group B was heavier than the other two groups,group A was also heavier than group C,all these had a statistical meaning(P<0.05).Normal side:there was no statistically significant among the all groups(P>0.05).3.Operated side:compared with the sham operation group,the MuRF-1expression of triceps surae of the model group and nerve mobilization group had a significant difference,which had a statistical significance(P<0.05);compared with the model group,the MuRF-1content of the nerve mobilization group A and B were obviously lower than the model group,the difference was statistical(P<0.05),while there were no significant differences between the group C and the model group(P>0.05);The compare among the three groups of the nerve mobilization,the content of group B was significantly lower than the other two groups,group A was also lower than group C,these were all statistically significant(P<0.05).Normal siderthere were no obvious difference among the groups(P>0.05).Conclusions1.The optimal stretching parameter of the nerve mobilization in this experiment is9%. 2.The nerve mobilization with the stretching parameters which limited in safe range can effectively delay the speed of muscle wet mass reduction after denervation.3. The nerve mobilization with the stretching parameters which limited in safe range can effectively reduce the Murf-1protein content.4.Reducing the expression of Murf-1protein may be one of the mechanisms which effectively delay the muscle atrophy after denervation by nerve mobilization.
