Background and aim:Chronic hepatitis B is one of the diseases which causeserious damage to human health.According to the world health organization carries the hepatitis B virus, there are350million people all over the world. About1million people die from HBV infection every year.Interferon alpha and nucleoside analogues (acid) can reduce the part of virus content with patients, but the interferon alpha efficient is only25%to40%, and the adverse reactions strictly limit the scope of its use.there is need to long-term medication, easy to relapse after drug withdrawal, easy to produce resistance shortcomings to nucleoside analogues (acid). More importantly, it is difficult to completely eliminate the hepatitis B virus, so it is necessary to actively explore new methods for the treatment of chronic hepatitis B.The idea is generally accepted that HBV not to take damage to the liver, hepatitis B virus itself after HBV infection mainly through the body’s immune response to the virus and cause liver cell damage.CD8+T cell mediated by HBV specific CTL response play a key role, is to start the liver cell damage and clearance of hepatitis B virus.The exploration to enhance HBV specific CTL response method is necessary, for the first task is to determine such research of HBV specific CTL response can be induced by antigen or with specific tables with a broad range of immunogenicity.Traditional Chinese medicine has a unique advantage in the treatment of chronic hepatitis B, treatment based on syndrome differentiation is the essence of traditional Chinese medicine, so it is necessary to explore HBV specific CTL response and the relationship between traditional Chinese medicine syndrome types, as to provide theoretical basis for combination of Chinese and western medicine therapy on chronic hepatitis B.Many studies have confirmed that there is a close relationship between the body’s immune and traditional Chinese medicine syndrome types of chronic hepatitis B patients, immune index detection provides objective basis for traditional Chinese medicine syndrome types, but the relationship between traditional Chinese medicine syndrome types and HBV specific CTL response induced by antigen or peptides in CHB patients have not been reported.Methods:60subjects with chronic hepatitis B were collected,according to type of traditional chinese medicine,30subjects to the syndrome of Liver and Gallbladder Wetness-heat type and other30subjects to the syndrome of hepatic stagnation and spleen deficiency type. PBMC of patients were sepatated from peripheral blood by Ficoll-Hypaque density gradient centrifagation and CTL response induced by HBcAg, HBsAg and mixed peptides were detected by Enyzme league immune spots (ELISPOT).Results:Comparied with CTL response induced by HBcAg, HBsAg and mixed peptides,HBV specific CTL response induced by HBcAg was significant strong (P<0.05). CTL response induced by antigen or mixed peptides with the syndrome of Liver and Gallbladder Wetness-heat type was better than the syndrome of hepatic stagnation and spleen deficiency type (P<0.05)Conclusion:These results suggest that HBcAg may be better antigen for inducing HBV specific CTL response and CHB with liver and the syndrome of Liver and Gallbladder Wetness-heat type may be received good therpation by induceing HBV specific CTL response.
