Objective:To observe the expression of associated TLRs and inflammatory chemokines of HT-29cells infected with human rotavirus (HRV) and intervention effect of Qiweibaizhu Powder.Methods:Select no polluting and good morphology of good HT-29cell lines that were infected with HRV、CoxB3、Shigella Flexneri and were randomly divided into model group, Qiweibaizhu scattered group, ribavirin group(HRV infection and CoxB3positive control), norfloxacin group (positive for Shigella infection Drug control), and normal control group,37℃5%CO2cultured48h, cultures were harvested. To observe the expression of TLR2,TLR3,TLR4,TLR7,TLR8mRNA and inflammatory chemokines IP-10, TNF-a, IL-8mRNA by RT-PCR technology.Results:1.The expression of TLRs on HT-29cell that infected with three enteropathogen:1) There was no significance difference between HRV model group and normal control group about TLR2mRNA expression level(P>0.05),but HRV model group TLR2mRNA expression were significantly lower than Shigella Flexneri model group(P<0.05)、CoxB3model group(P<0.05).2) HRV model group TLR3mRNA expression were significantly higher than CoxB3model group(.P<0.05)、Shigella Flexneri model group(P<0.05)、normal control group(p<0.05).There was no significance between CoxB3and normal control group(P>0.05)、Shigella Flexneri group and normal control group(P>0.05).3) HRV model group TLR4mRNA expression were significantly higher han normal control group(P<0.05)、lower than CoxB3model group(.P<0.05)、Shigella Flexneri model group(P<0.05).The expression of TLR4in Shigella Flexneri model group had increased, and there was significantly difference between shigella Flexneri model group and HRV group、CoxB3group(P<0.05).4) HRV model group TLR7、TLR8mRNA expression were significantly higher than normal control group(P<0.05),and higher than CoxB3model group(P<0.05)、Shigella model group(P<0.05).2.The intervention effect of Qiweibaizhu Powder:1) the expression of TLR3、TLR7、TLR8mRNA in the HRV+Qiweibaizhu Powder group were lower than the model group(P<0.05),Compared with normal control group,the expression of TLR3、TLR7、TLR8in the HRV+Qiweibaizhu Powder group totally increased(P<0.05),there was no significantly difference between HRV+Qiweibaizhu Powder and ribavirin group(P>0.05);Compared with the model group,the expression of TLR7、TLR8mRNA in the CoxB3+Qiweibaizhu Powder group totally decreased(P<0.05),the expression of TLR7、TLR8mRNA in the HRV+Qiweibaizhu Powder group were higher than normal control group(P<0.05),compared with ribavirin group, the expression of TLR3、TLR7mRNA in the HRV+Qiweibaizhu Powder group have no significantly difference(P>0.05),the expression of TLR8mRNA increased(P<0.05).2) Compared with normal control group,model group IP-10,TNF-a,IL-8mRNA expression were significantly higher(P<0.05); there was significantly difference between model group and Qiweibaizhu scattered group(P<0.05);and there was no significantly difference between Qiweibaizhu scattered group and ribavirin group (P>0.05).Conclusion:1.After HT-29cell infected with HRV,TLR2、TLR4cann’t recognise HRV dsRNA.2.After HT-29cell infected with HRV,TLR3、TLR7、 TLR8in the HT-29cell all increased.3.Qiweibaizhu Powder can not prevent the function that TLR3、TLR7and TLR8recognise HRV dsRNA,and cann’t completely prevent the response to HRV of TLR7、TLR8.4.Qiweibaizhu Powder can relieve the inflammation pathological reaction that caused by HRV through making the expression level of IP-10mRNA、 TNF-amRNA、 IL-8mRNA down-regulate.
