Objective: Polymorphisms in angiotensin II type-1/2receptor (AGTR1/AGTR2) genes may be involved in the pathogenesis of primary aldosteronism (PA). The present study aims to reveal some loci susceptible to the disease on the genes of angiotensin â…ˇ receptors.Methods: A case-control study was conducted on202patients diagnosed with primary aldosteronism and188controls. Clinical data and whole blood of all subjects were collected and the leukocyte genomic DNA was extracted. Ten tagging SNPs on AGTR1/AGTR2were genotyped for all subjects via the method of improved multiplex PCR-ligase detection reaction. Statistical analysis was performed with chi-square test and logistic regression analyses using the SPSS13.0. A web-based program SHEsis and Haploview4.2were applied to manage the calculation of linkage disequilibrium and Hardy-Weinberg equilibrium.Results: Nine out of ten studied SNPs have genotype distributions compatible with the Hardy-einberg equilibrium in both cases and controls (p>0.05). The rs3772616C allele on AGTR1gene was a susceptible factor for PA (p<0.001) and the TT genotype significantly decreased the risk of PA compared to CC homozygote (p=0.008, adjusted OR=0.13;95%CI:0.03-0.59). The polymorphism of rs3772616was associated with PA under the additive and dominant models. The female carriers of G allele in rs5193showed significant difference compared to T allele. No significant difference was observed in the distributions of haplotypes for two genes between two groups.Conclusions: The AGTR1rs3772616can be considered as a hereditary marker for PA. The rs5193polymorphisms of AGTR2gene may be associated with the predisposition to PA for women.
