Objective To investigate the effect of oncolytic adenovirus (Ads) combined with radiation therapy in bladder tumor cells.Methods The cell biability was determined by CCK-8(cell counting kit-8) assay. Cell revolution and apoptosis were analyzed by flow cytometry. The mRNA expression of Bax, Bcl-2、 BECN、Atg-5、CAR were analyzed by realtime-PCR. The protein level of E1A, Bax, Bcl-2, Cytochromin c were measured by Western blot.Results Oncolytic adenovirus combined with radiation therapy significantly inhibited the proliferation of bladder tumor cells in an concentration-dependent manner, indicating a synergetic effect. Flow cytometry showed that combination treatment could improve the apoptosis rate of bladder tumor cells. The mRNA expression of Bax, CAR was up-regulated and the mRNA expression of Bcl-2was down-regulated (p<0.05). Compared with the contral group, the autophagy related gene BECN、Atg-5mRNA were down-regulated (p<0.05).The protein level of E1A,Bax, Cytochromin c (cytoplasma) were up-regulated and the protein level of Bcl-2, Cytochromin c (mitochomdria) were down-regulated by combination treatment.Conclusion Oncolytic adenovirus combined with radiation therapy exerts its antitumor effect through inducing mitochondrial apoptosis pathway and had a synergetic effect.
