Objective To investigate the apoptosis effects and related expressions of HERGpotassium channel of human glioma U251cell lines induced by berberine hydrochloride.Methods in vitro,use different concentrations of berberine hydrochloride to intervene inhuman glioma U251cells, determined by Methylthiazolyltetrazolium (MTT) was used toevaluate the cellular proliferation inhibition rate;Hoechst33258staining with fluorescentmicroscope observe whether the cell morphology have changes of apoptosis or not;applyflow cytometry (FCM) to detect the cell apoptotic rate and distribution of apoptotic rate ondifferents of stages; apply real-time fluorescent quantitative reverse transcriptionPCR(RT-PCR) to detect mRNA of HERG and apply western blotting(WB) to detectprotein expression of it in order to observe whether this changes with dose of drug or not;Results Determined by the MTT method result showed that the cellular inhibitionrate was significantly increased with concentrations of berberine hydrochloride,showed on dose and time-dependent relationship, the the half maximal inhibitory concentration(IC50) was8.78±0.20μmol/l for24h. Hoechst33258fluorescent stainned examine revealed that cell morphology of treated group displayed the typical characteristics of apoptosis, the early apoptotic rate and late apoptotic rate raised as dose ofit by FCM. The both mRNA and protein expression of HERG which were detectedrespectively by RT-PCR and WB,was increasingly increased with dose of drug.Conclusion: Berberine hydrochloride could induced apoptosis of U251cell in vitro,furthermore regulated up protein expression of HERG potassium to inhibit U251of thecellular proliferation.
