Objective: With soft liver strengthen spleen, activating blood stasis methodto guide the compatibility of medicines, discuss the application of the law indrug-induced liver injury. By experimental observation on the liver enzyme reductionparty CCL4induced liver injury in mice liver function index of ALT, AST,pathological changes and the effect of hepatic CYP2E1expression, liver spleen,invigorate the circulation of blood stasis was explored for the therapeutic effect ofdrug-induced liver injury and its mechanism, for liver enzyme reduction in laying afoundation for the clinical application of drug-induced liver injury.Methods: Choose6weeks of age60male kunming mice, adaptability to feeda week later, the points and six groups at random, in the order: normal group(n=10),model group (n=10), western medicine group (n=10), Chinese traditional medicinehigh dose group(n=10), estrogen group (n=10), low dose group (n=10). Using20%carbon tetrachloride olive oil, according to the5ml/kg, intraperitoneal injection,2times a week, four weeks later, began to fill the stomach, the normal group, modelgroup by gastric volume of physiological saline, continuous lavage4weeks. Afterfeeding, at the end of the fasting water for24hours, put to death in mice, eyeballblood, to detect the contents of serum ALT, AST. Liver tissue pathological observation.Using immunohistochemical staining method detecting hepatic CYP2E1expression.Data through the SPSS statistical software, differences between groups compared byT test, all the data said by x s.Results:(1)Serum transaminase test results: compared with blank group ofmice serum ALT, AST, content of model group were significantly increased, withstatistical significance (P <0.01), said building success; Western medicine andChinese medicine group serum ALT, AST, significantly lower than the model groupwas statistically significant (P <0.05); Traditional Chinese medicine (TCM) of serumALT, AST, high dose group was obviously lower than western medicine group (P <0.05); Low dose group of ALT and AST decreased, but no statistical significance compared with model group (p st130.05). Comparison between traditional Chinesemedicine way, serum ALT, AST dose group was obviously lower in the low dosegroup (P <0.01, P <0.05), serum ALT, AST, high dose group was obviously lowerdose group (P <0.01).(2)Liver tissue pathological observation: a wide range of livercell swelling, massive hemorrhage stove, visible bleeding around the liver celldegeneration, necrosis, inflammatory cell infiltration within portal area and lobular.Compared with model group, western medicine and Chinese medicine high, middledose group have different degrees of improvement, with traditional Chinese medicineto improve the effect of high dose group is better, low dose group compared withmodel group have no obvious change.(3)CYP2E1immunohistochemical stainingresults: each group mice were positive expression of different level. Positive ratecomparison, constituting the model of CYP2E1positive rate is greater than thenormal group, the difference was statistically significant (P <0.01); Western medicineand Chinese medicine group CYP2E1positive rate is less than the model controlgroup, the difference was statistically significant (P <0.01); Chinese medicine highdose group of positive rate compared with the western medicine group, with statisticalsignificance (P <0.05).Conclusion: Mice induced by CCL4in liver damage, liver function index ofALT and AST increased significantly, liver cell swelling, a massive hemorrhage stove,degeneration, necrosis, inflammatory infiltrates, objectively reflect the drug-inducedliver injury model. Chinese medicine liver enzyme reduction can reduce abnormalliver function index in mice after liver damage, reduce serum transaminase, improvethe pathological changes of the corresponding, the prescription by influencing theliver tissue of CYP2E1expression, achieve the goal of treatment of drug-inducedliver injury, lay the foundation for its clinical application.
