Objective To investigate the effect of the extracts of4’-demethyl-4-deoxypodop hyllotoxin and4’-demethyl-4’-oxygen-cyclopropane carbonyl group-4-deoxypodophylloto xin on inhibiting proliferation A549、HepG2、MG-63、HeLa cells in vitro; To explor e the acute toxicity of the4’-demethyl-4-deoxypodophyllotoxin and Anthriscus lacton e’s Effect of expression on STAT3, P53, NF-κBP65protein.Methods Research the inhibition of different concentration4’-demethyl-4-deoxypodoph yllotoxin and4’-demethyl-4’-oxygen-cyclopropane carbonyl group-4-deoxypodophyllotox in in the A549、HepG2、MG-63、HeLa cells by the method MTT. Calculate the rate and of the drug on the inhibition of different cells and reckon the IC50accordingly.O bserve the reaction of the mices by the dosage regimen of2000mg/kg,0.4ml/10g4’-de methyl-4-deoxypodophyllotoxin.Compare the effect of expression on STAT3, P53, NF-κBP65protein with40μg/ml Anthriscus lactone group and1μg/ml Paclitaxel group.Results1.4’-demethyl-4-deoxypodophylIotoxin showed strong inhibition of prolif eration on A549, HepG2, MG-63, HeLa cells.The IC50are45.29μg/ml、66.36μg/ml、57.89μg/ml、35.58μg/ml.4’-demethyl-4’-oxygen-cyclopropane carbonyl group-4-deox ypodophyllotoxin showed strong inhibition of proliferation.too.The IC50are47.73μg/ml、53.57μg/ml、38.29μg/ml、91.49μg/ml.The Inhibition effect of these two derivatives on the proliferation showed no difference (P>0.05)2.3of21tested mices were dead.The cause of death was intragastric damage.The s urvival mices return to normal in7days. We can Speculate the toxicity of4’-demet hyl-4-deoxypodophyllotoxin is not strong.3. Compared with the control group,expression on STAT3, NF-κBP65protein was re duced.Expression on P53protein was raised in the Anthriscus lactone group and Pacl itaxel group.Conclusions4’-demethyl-4-deoxypodophyllotoxin and4’-demethyl-4’-oxygen-cy clopropane carbonyl group-4-deoxypodophyllotoxin can inhibit the proliferation of A549、HepG2、MG-63、HeLa cells. The toxicity of4’-demethyl-4-deoxypodophyllotoxin i s not strong.The inhibition may be achieved by reducing the expression on STAT3. NF-κBP65protein and raising the expression on P53protein.
