Objective: To establish an animal model of periodontitis in male wistarrats by ligation with periodontal method, observed diacerein therapeuticeffects of periodontitis in rats, for diacerein as a result of systemic drugtreatment of periodontitis provide the experimental basis.Methods:3-month-old purebred male wistar rats were randomly dividedinto four groups: normal group10, periodontitis10, four weeks treatmentgroup10,8weeks treatment group10.Periodontitis and treatment group at10%chloral hydrate abdominal cavity (350mg/kg) anesthesia,were ligatedbilaterally of their first maxillary molar teeth with0.2mm steel-wire;theligature wire inside the free gingival,not damage the junction epithelium,highsugar water instead of drinking water on the ligation of the day.After1monthof periodontal ligation,group Normal and group periodontitis were randomlyselected and killed,take the rats’maxillary,make maxillary first molar toothperiodontal united biopsy was processed for histological observation,toidentify periodontal model.Periodontitis model was established,drug groupwere treated with diacerein,according to the proportion of100mg/kg bodyweight2ml daily gavage,periodontitis group were treated with nature salt2mldaily.After4weeks and8weeks medication,rats were bled by femoral arteriesbledding,the rats maxillary were immediately putted into10%formaldehydebuffer solution for fixation,then by10%EDTA decalcification solution,theproduction of dental week biopsy were processed for HE staining toobserve,light microscope observation; Tartate resistant acidphosphatase(TRAP) staining to observe osteoclast’s quantity anddistribution,and make statistics analysis processing; Immunohistochemistywas used to observe Interleukin-1β (IL-1β) the expression and makes statisticsanalysis processing. Result:1After periodontal ligature, periodontitis group and treatment group ratsloss of appetite, poor mental state, reduce body weight than normal group;Oneweek after the building, the rat diet gradually returned to normal, good mentalstate. After the successful model of periodontitis, periodontitis group and thetreatment group were given normal saline、 diacerein respectively,themedication rats appeared mild symptoms of diarrhea, increase of urine, urinecolor become more yellow, eat less, spirit is weak, a week after the diarrheasymptoms, spirit gradually returned to normal. Gavage after8weeks, the ratsweight has increased, most normal group, periodontitis group is minimal.24weeks after modeling,take the maxillary, macroscopic observation,the normal group had no significant inflammation, no bleeding on probing;periodontitis group showed swollen gums, bleeding on probing, gum and toothsurface does not fit, food residue debris accumulation, the formation ofshallow periodontal pockets. Make maxillary first molar tooth periodontalshow that normal group periodontal structural integrity, no inflammation.Periodontitis group of periodontal inflammation is significantly, gingivalepithelium contains a large number of inflammatory cells infiltration,combination of epithelium and root is not close, on root proliferation or shift,the periodontal membrane expansion of capillary hemorrhage, alveolar ridgecap with a small amount of absorption.Periodontitis model set upsuccessfully.3HE stained histological examination of periodontal tissue: NormalGroup: junction epithelium located at the cemento-enamel junction, noinflammatory cell infiltration, periodontal ligament fibers arranged in neatrows, the alveolar crest without significant absorption. Periodontitis group: acombination of epithelial proliferation or shift to the root square, surroundedby a large number of inflammatory cell infiltration, formation of deepperiodontal pockets, telangiectasia bleeding, the gap widened periodontalligament, periodontal ligament fibers derangement, alveolar crest absorbsignificantly.4weeks treatment groups: periodontal pocket lighter, still belowthe epithelial and inflammatory cell infiltration, alveolar crest see a small amount of osteoblasts, see the alveolar crest around a small osteoclasts.8weeks treatment groups: periodontal pocket lighter, alveolar crest see a lot ofosteoblasts form new bone.4Tartrate-resistant acid phosphatase dyeing determines osteoclast’sdistribution and the expression level in the periodontal tissues of rat, NormalGroup: osteoclast can be seen in the alveolar crest area, only individualosteoclast existence in the alveolar crest edge.Periodontitis group: in thealveolar crest area massive osteolclasts existence.4weeks treatment groups: inthe alveolar crest edge osteoclasts are few.8weeks treatment groups: in thealveolar crest area osteoclasts are rare.The alveolar crest area osteoclastcounting results,periondontitis group was higher than the normal group andtreatment groups(P<0.01) significantly,8weeks treatment group and normalgroup had no significant difference(P>0.05),8weeks group was significantlylower than4weeks group(P<0.01).5Immunohistochemistry determination of IL-1β distribution ofexpression in the periodontal tissues of rat’s: mainly located in the epithelialcells, neutrophils, monocytes-macrophages, vascular endothelial cells,fibroblasts and other cells cytoplasm and matrix. IL-lβ in the normal group ofperiodontal tissues showed weak and positive expression. Periodontitis groupin inflammatory cells infiltration and fibroblasts was strongly positiveexpression of lesion.4weeks treatment group positive cell number decreased,8weeks treatment group positive cell number decreased obviously, and in thefibroblasts assumes the weak positive expression.IL-1β average light densityvalue determination result, periodontitis group obviously was higher than theother groups (P<0.01), between the normal group and8weeks treatment groupwas no obvious difference(P>0.05),4weeks treatment group was higher than8weeks treatment group(P<0.01).Conclusion:1By ligation of the maxillary first molar successfully established a ratmodel of periodontitis.2Diacerein capsules significantly reduced the formation of periodontal tissues of rats osteoclasts, reducing the expression of IL-1β, reducing theperiodontal local inflammation,induces the bone, promoting the teeth alveolarbone formation of new bone, have a therapeutic effect on periodontitis.
