Objective:Renal cell carcinoma(RCC) stem from renal tubularepithelial cells,which accounts for80%to90%of renal malignanttumor.There are about more than20million people which are diagnosedin kidney cancer each year in the world. Incidence of renal cell carcinomais just below to the bladder cancer in malignant tumor of urinary systemin our country.Therefore, It is very vital significant to explore thepathogenesis of renal cell carcinoma, and find new therapeutic targets andeffective drug treatment for human.In recent years, it has become a hotspot that between the Wnt signaling pathway and the tumor.Abnormalactivation of the Wnt signaling pathway in the factor may cause cellmalignant change in study. DKK-1and DKK-3gene belong to the tumorsuppressor genes in the Wnt signaling pathway and the mechanism ofwhich has not been clear in renal cell carcinoma.To analyze the role ofthe process of formation, progression and metastasis in renal cellcarcinoma, reverse transcription polymerase chain reaction(RT-PCR) andimmunohistochemistry was used to investigate the expression of DKK-1and DKK-3in renal cell carcinoma. The research was also designed topresent a potential application in the treatment of renal cell carcinoma.Methods:1Reverse transcription polymerase chain reaction (RT-PCR) methodwas used to detect the mRNA level of DKK-1and DKK-3gene in36cases of RCC tumor tissues and normal tissues.2Immunohistochemistry was used to investigate the proteinexpression of DKK-1and DKK-3in47cases of RCC and31cases ofnormal tissues;3SPSS13.0was used to analyze the data of the experiment. Results:1The relationship between expression of mRNA and protein ofDKK-1and DKK-3and RCC clinical pathology1.1The mRNA expression of DKK-1and DKK-3in RCCDKK-1mRNA expression in tumor tissues (0.21±0.06) wassignificantly lower than that in normal tissues (0.28±0.05)(P<0.05).DKK-3mRNA expression in tumor tissues (0.65±0.15) was significantlylower than that in normal tissues (0.79±0.13),the difference wasstatistically significant(P<0.05).1.2The protein expression of DKK-1and DKK-3in RCCThe positive expression rate of DKK-1in RCC (59.5%) wassignificantly lower than that in normal tissues (90.3%)(P<0.05). As therise of TNM stages,the positive expression rate of DKK-1reduced.Thepositive rate was higher in the group which the tumor diameter≤7.0cmthan that in the group which the tumor diameter>7.0cm. The positiverate in the group of lymph node metastasis and lymph nodenon-metastasis was10.3%(1/14)and66.8%(22/33)respectively, thedifference was statistically significant(P<0.05), whereas the expression ofDKK-1was correlated with neither gender nor age,the difference was notstatistically significant(P>0.05).The positive expression rate of DKK-3in RCC (65.9%) wassignificantly lower than that in normal tissues (93.5%)(P<0.05). But Ⅱperiod compared with Ⅲ+Ⅳ stage was not statisticallysignificant(P>0.05).The positive rate was higher in the group which thetumor diameter≤7.0cm than that in the group which the tumor diameter>7.0cm. The positive rate in the group of lymph node metastasis andlymph node non-metastasis was21.4%(3/14)and60.6%(20/33)respectively,the difference was statistically significant(P<0.05), whereasthe expression of DKK-3was correlated with neither gender nor age,thedifference was not statistically significant(P>0.05).2There is no ralativeship between the expression of DKK-1and DKK-3.Conclusions:1The expression of DKK-1and DKK-3in RCC is significantlylower than that in normal tissues, which suggests that the reduction ofDKK-1and DKK-3may be involved in the formation of RCC and usedas an index of prognosis of renal cell carcinoma.2The expression of DKK-1and DKK-3is correlated with TNMstages,lymph node metastasis and tumor diameter,the positive rate washigher in the group of lymph node metastasis than that in the group oflymph node non-metastasis,the positive rate was higher in the groupwhich the tumor diameter≤7.0cm than that in the group which the tumordiameter>7.0cm,but not correlated with neither gender nor age, As therise of TNM, their positive expression rate of DKK-1reduced whichsuggests that the down-regulation of DKK-1and DKK-3may becontributed to progression, infiltration and invasion of RCC.3There is no ralativeship between the expression of DKK-1andDKK-3.
