Objective:Renal cell carcinoma (RCC) is one of the most commonmalignant tumor in urinary system. Its incidence and mortality havecontinuously increased during the last50years. Recent studies showed thatthe incidence of RCC was higher than that of bladder cancer in somewhere,and increased by2%per year. Nearly half of RCC patients were at advancedstage or metastatic at the first consult. Thus, early diagnosis and interventionof RCC at the genetic level are key points in the further studies.In recent years, many studies have shown that the process of cellsmalignant transformation involves multiple complex signal transductionpathways. Cyr61, rich Cysteine protein61(Cysteine rich CCN1/Cyr61) wasdiscovered first, and was the most representative of the CCN protein familymembers. It is secreted, extracellular matrix proteins, involved in regulating avariety of biological processes of cells, such as differentiation, metastasis,angiogenesis, cell adhesion, proliferation, survival, and apoptosis. Cyr61expresses in multiple types in tumor tissues, and plays an important role in theprocess of tumor formation.This experiment was to study the expression of CCN1/Cyr61in normalkidney tissue. Investigate the role of Cyr61/CCN1in the development ofkidney cancer, in order to provide reference for clinical diagnosis of renalcancer, and to provide new targets for kidney cancer targeted therapy.Methods:RT-PCR method was adopted to detect the transcription ofCCN1in22cases of normal kidney tissue and48cases of RCC tissue.Immunohistochemistry method was adopted to detect the expression of CCN1in22cases of normal kidney tissue and48cases of RCC tissue. We analyzedthe role of Cyr61/CCN1in the occurrence and development of kidneycancer. Results:1The result of RT-PCR showed that relative transcription genesof CCN1/Cyr61in kidney cancer group is significantly higher than normalkidney, and the differences between groups was statistically significant((P<0.05).2Immunohistochemical result showed that the positive expression rate ofexpression rate of CCN1/Cyr61:72.9%(35/48) was higher than the controlgroup:27.3%(6/22), and the difference between groups was statisticallysignificant (P<0.05).3Expression of Cyr61/CCN1levels in the early-stage renal carcinomatissue:63.3%(18/30) was higher than those in normal kidney tissue:27.3%(6/22), and the difference showed significant statisticalsignificance(P<0.05).4CCN1expression level in the early renal cancer:63.3%(18/30) waslower than those in advanced kidney cancer:94.4%(17/18), the differenceshowed statistical significance(P<0.05).5CCN1expression level is higher in the metastatic kidney tissue:100%(9/9) than that in the non-metastatic kidney tissue:66.7%(26/39), thedifference is statistically significant(P<0.05).Conclusion:1The expression level of CCN1in RCC is higher than thatin normal renal tissues, CCN1, as the cancer gene, plays an important role inthe formation of RCC.2In the early-stage of RCC, the expression level of CCN1was higherthan that in normal kidney tissues, and the result showed statisticallysignificant difference, which suggests that CCN1may be a specific indicatorin early diagnosis of renal cancer.3Expression level of CCN1in early-stage of RCC was lower than that atadvanced stage, which suggests that CCN1plays an important role in thedevelopment of the RCC.4Expression level of CCN1in tissue of RCC was higher than that innormal renal tissue, CCN1may play an important role in the occurrence ofrenal cancer, by activating of multiple signaling pathways, and regulating the formation of new blood vessels. CCN1may become the target of geneticintervention.
