HER-2is a kind of epidermal growth factor receptor, plays animportant role in the development of gastric cancer. Studies have shownthat, HER-2positive expression of gastric cancer has its own peculiarities,and exhibited at the molecular targeted therapy good effect. ResearchHER-2expression in gastric cancer and gastric cancer to analyze itsrelationship with the biological significance, providing new therapeuticapproaches for the clinical treatment of gastric cancer.Purpose: Human epidermal growth factor receptor (Human epidermalgrowth factor receptor-2HER-2) expression in patients with gastric cancerdetection, analysis of their expression and biological behavior and clinicalcorrelation. Comparison of different methods to detect pre-HER-2resultsin consistency, comparing the results of HER-2postoperative consistencyin gastric cancer heterogeneity and IHC methods to determine the effect ofHER-2process quality control of HER-2expression.Methods: Real-time quantitative PCR (Realtime Polymerase ChainReaction, Realtime PCR) measured HER-2gene expression in142cases ofgastric carcinoma before surgery, by immunohistochemistry(Immunohistochemistry, IHC) measured before surgery and96cases of277cases of gastric carcinoma postoperative gastric tissue hER-2proteinexpression analysis with sex, age, tumor location, Lauren classification, thedegree of differentiation of patients, depth of invasion, lymph nodemetastasis, distant metastasis, TNM stage, vascular invasion, invasion ofnerves correlation. Comparison of different methods to detect theconsistency of the results of HER-2, compared to the same patients beforesurgery to detect tissue after surgery IHC HER-2protein expressionconsistent results, and to explore the heterogeneity of gastric cancer HER-2 IHC method for the determination of the quality control process for theHER-2protein expression.Results:194cases of gastric carcinoma before surgery through the IHCmethod measured HER-2protein overexpression rate11.702%;2277cases of gastric carcinoma postoperatively measured by IHCHER-2protein expression was7.581%;3142cases of gastric carcinoma before surgery through RealtimePCR method measured HER-2gene amplification rate of12.676%;4Compared to the previous one gastric carcinoma after surgeryIHC methods were applied to detect HER-2protein over-expression was nosignificant difference (t=0.948, P=0.349>0.05);5Compared to the previous application of the same organizationgastric surgery Realtime PCR method measured with the HER-2geneamplification was detected after application of IHC HER-2proteinoverexpression was no statistically significant difference (t=-1.346, P=0.186>0.05);6HER-2over-expression in gastric cancer and the patient’s age,sex, tumor location, Lauren classification, vascular invasion, neuralinvasion was no significant difference (P>0.05), while patients with thedegree of differentiation, depth of invasion, lymph node metastasis, distantmetastases, TNM staging a significant difference (P <0.05);7Improved detection technology through the front IHC HER-2protein expression was5.000%, the improved HER-2protein expressionwas8.017%;8Multi-center measured gastric HER-2protein overexpression was100%concordance rate.Conclusions:1Gastric HER-2protein overexpression correlated with age, sex,tumor location, Lauren classification, vascular invasion, nerve invasionwas no significant difference (P>0.05), and with the depth of invasion, lymph node metastasis, distant metastasis, TNM stage increase, HER-2protein overexpression rate increases, and HER-2protein over-expressionof a large proportion of the share in the differentiation of gastric cancer;2Preoperative and postoperative after IHC method to detectHER-2protein overexpression results are consistent;3Realtime PCR and IHC methods to detect HER-2expressionconsistent with the results;4Strict quality control process to detect the expression of HER-2test results can be significantly improved;5Gastric tumor heterogeneity under strict quality control, the lessaffected by the IHC method to detect HER-2protein expression in gastriccancer, the measured result is true and reliable.
