Objective: To explore the nursing intervention for Alzheimer’s patientsand provide objective experimental basis through observing the effect aboutmusic therapy and exercise therapy on learning and memory abilities andcholine acetyltransferase in hippocampal synapse in SAMP8mice.Methods: Forty healthy male seven moon’s age SAMP8mice, whichweight of28~32g, breeding in normal temperature, light, free drinking water,feeding conditions were randomly divided into P8control group (n=10),musicgroup (n=10),exercise group (n=10),music and exercise group (n=10).Tenhealthy male SAMR1mice as homology normal control(to call for short Rgroup).The music group and music and exercise group were broadcastedmusic at8:00~9:00and20:00~21:00every day,kept for6weeks and6day perweek. The track was given priority to with cheerful music,such as PhoenixLegend’s Coolest Ethnic and Flying Free,etc. Every time listening to themusic to fix mice in a quiet room, shut down windows and doors, pull acurtain, fixed position every time, the sound (songs sound lang GL-02) awayfrom the cage of distance of1.5m, db is70~80db.The exercise group andmusic and exercise group were kept for swimming at15:00every day for6weeks (once a day,6day per week). Every time swimming for15minutes.The pool size is100cm in diameter and50cm deep, water temperature is28℃plus or minus2℃and room temperature is26℃, the depth of water is40cm. Manual control mice swimming, to prevent drowning deaths, whenmice to exhaustion, it takes out from inside the pool, after3minutes rest, andthen put it in the swimming pool, swam enough provisions of swimming timeevery day.Postexercise6weeks, to evaluate SAMP8mice’s learning andmemory abilities by Morris water maze Place navigation test and Spatial probe test. After Morris water maze, the paraformaldehyde-perfused brainswere obtained or the hippocampus tissue proteins were divided on icewaterbath, to respectively investigate the expression of ChAT in hippocampus inSAMP8mice by immunohistochemical techniques and western-blot method.All relults demonstrated with x±s are analyzed by SPSS19.0, P<0.05denotesignificance.Results:1Morris water maze test results: The escape latency is affected by timeand group interaction. The first day all groups escape latency is No statisticalsignificance (P>0.05). The second day and third day, compared with othersgroups, P8control group escape latency is the longest (P<0.05), the othersgroups is No statistical significance (P>0.05). The fourth day and fifth day, P8control group escape latency is the longest (P<0.05), R group escape latencyis the shortest (P<0.05), the others groups is No statistical significance(P>0.05). In spatial probe test, compared with others groups, P8control groupfrequencies of crossover platform are the least (P<0.05), R group frequenciesof crossover platform are the most (P<0.05), the others groups is No statisticalsignificance (P>0.05). There is no interaction in music therapy and exercisetherapy(P>0.05).2Immunohistochemical results: compared with others groups, P8controlgroup average optical value of ChAT protein is the minimum (P<0.05), Rgroup average optical value of ChAT protein is the maxlmum (P<0.05).Thereis a little difference with the others groups average optical value of ChATprotein, but is No statistical significance (P>0.05). There is no interaction inmusic therapy and exercise therapy(P>0.05).3Western-Blot results: compared with others groups, P8control groupexpression dose of ChAT protein is the minimum (P<0.05), R groupexpression dose of ChAT protein is the maxlmum (P<0.05), both of musicgroup and exercise group is less than music and exercise group (P<0.05).There is a interaction in music therapy and exercise therapy (P<0.05). Conclusion:1Music therapy and exercise therapy can improve the learning andmemory abilities of SAMP8mice and increase the content of the ChAT inhippocampus.2Music therapy and exercise therapy improve the learning and memoryability of SAMP8mice may be related to improve the content of thehippocampus ChAT.3Music therapy and exercise therapy can improve the content of theChAT in hippocampus of SAMP8mice, but compared with the normal ageSAMR1mice, there are still differences in ChAT content.4If music therapy and exercise therapy combined action is moreeffective than monotherapy, still need further study.5Music therapy and exercise therapy applied to alzheimer’s patient care,their specific treatment still need further discussion.
