Objective:To investigate drug resistance in Acinetobacter Baumannii tocommonly used antimicrobial drugs. To detect the efflux pump systematicphenotype and genotype of the panresistant Acinetobacter baumannii.Toinvestigate the function and gene expression of acinetobacter baumannii activeefflux pump in the resistance. To improve effciency of antibiotics, therapeuticeffect of Pan-resistant Acinetobacter baumannii, prognosis of patients,andprovide theoretical basis for the research and development of new antibacterialdrugs.Methods:1A total of142non-duplicate clinical isolates of Acinetobacterbaumannii were collected. Minimum inhibitory concentration (MIC)of19kinds of antimicrobial drugs were detected for Acinetobacter baumannii:amikacin, gentamicin, ampicillin, ampicillin/sulbactam, piperacillin,ceftazidime, imipenem, cyproterone gatifloxacin, cefoperazone/sulbactam,piperacillin/tazobactam, meropenem.2The impact of Carbonyl cyanide chlorobenzene hydrazone (CCCP) onthe MIC of strains:there are two kinds of agar medium,one is Antibioticdilution medium with pump inhibitors,another is medium only with pumpinhibitors.3To detect and analyze the efflux pump protein gene of externaldrainage pump positive strains: the DNA genes of the efflux pump phenotypicpositive strains were extracted by Maxwell16Cell LEV DNA PurificationKit, amplified by PCR. The electrophoresis were analyzed by gel ima-ging and analysis system. PCR products were selected to sequenced.4All results were analyzed with WHONET and SPSS l3.0.Results: 1The142clinical Acinetobacter Baumannii isolates were obtainedmainly from sputum (72.6%) and secretions(11.3%). Most isolates weresubmit by ICU (40.8%) and respiratory medicine (14.8%).2142strains were determinated for drug-sensitive. The sensitivity forolymyxin B was100%.The highest resistance rate was tetracycline(94.5%).The drug resistance rates of Norfloxacin, gentamycin, cefotaxime,ampicillin were more than90%. The resistance rates of imipenem andMeropenem were34.5%and36.6%. Among these isolates,96strains were.3After treated96multiple drug resistant and pan drug resistance strainswith CCCP, the resistance rate of gentamicin reduced from98.96%to33.33%,reduced63isolates of the drug resistant bacteria. MICs of32strains,41strains,29strains descended to1/4or less original MIC values withfor Tetracycline, norfloxacin, cefotaxime. MICs of only34strains descendedto1/4original MIC values with the above four antibotics.4Of the34isolates, the gene of adeB,adeR,adeS,adeJ,adeE,abeM wereidentified in33(97.06%),32(94.12%),33(97.06%),33(97.06%),0,33(97.06%)strains of bacteria,respectively.Conclusions:1Clinical isolates of Acinetobacter baumannii exists widely resistant tocommonly used antimicrobial agents.2After treated96multiple drug resistant and pan drug resistanceAcinetobacter baumannii strains with CCCP,MICs of only34strainsdescended to1/4or less original MIC values with four antibotics.3The adeB, adeR,adeS,adeJ,abeM gene were detected in34drugresistant strains suspected efflux pump protein Acinetobacter Baumannii.4adeB, adeR,adeS,adeJ,abeM gene were closed to that of referencestrains in GenBank with nucleotide homology100%.
