Cancer is one of the greatest enemies for human health. Radiotherapy, due to its wide indications and selectivity, will be applied to more than70%of malignancy patients at a certain stage of their treatment. The aim of radiotherapy is focus on the maximum radiological dose on the lesion area to exterminate tumor cells, while protect the normal tissues or organs need special protection. Because of normal tissues and tumors usually intertwined, neither traditional radiotherapy technology nor modern precise radiotherapy techniques can entirely protect normal tissues when exterminate tumor cells. Therefore, to find some methods to protect normal skin, tissues and organs as far as possible become the key to relieve patients’pain and improve the effect of treatment in radiotherapy. Here, we expressed and purified human cryptochrome1, a protein with the activity to absorb radiation, and detected its protective effects through cell based assays. We constructed prokaryotic expression vector pET28a(+)-hCRYl and expressed the recombinant protein in Escherichia coli BL21(DE3). The hCRY1protein was purified with high purity and its radiation protective activity was confirmed in Hela and HaCaT cell lines. Our results suggested a possible application of recombinant hCRY1in produce skin protective agent for radiotherapy.
