Group A rotavirus(RV) is the major pathogen of diarrhea disease in infants, which responsible for nearly600,000deaths per year worldwide. There is no effective medicine to treat rotavirus infection by now, and the protective efficiency of existing vaccine are very different in varied areas. In addition, the existing vaccine are expensive and having the potential safety problems, it is a urgen matter to develop a new vaccine which is safer, cheaper and more efficient. The reliable animal model is the guarantee of vaccine development, and helps understand the process of virus infection and pathogenesis. In this study, we tried to establish a stable rotavirus sucking mouse model, control affecting factors and evaluative indicators, and perform a preliminary evaluation. This research will provide effective technology tools for the development of rotavirus vaccine, therapeutic antibody and antiviral drug.Firstly, in this study, we developed a reliable panel of rotavirus for animal challenge. Then we successfully established the rotavirus sucking mouse model. With rotavirus attacking the7days-old sucking mouse, a series of changes could be observed, such as the signs of illness, slower rate of weight growth, diarrhea, pathological changes of tissues. Secondly, we used the sucking mouse model to perform a preliminary evaluation. Through acquired from dams or immune serum by intraperitoneal injection, antibodies could be effectively passed to the sucking mouse, and protected sucking mouse from diarrhea, reduced the damage of the tissue, inhibited virus replication. Then we analyzed the influence factors, advantages and disadvantages of each evaluation index, it helped to improve the stability and accuracy of the animal model.In summary, this study successfully established an rotavirus sucking mouse model, and make a preliminary evaluation for protection. This rotavirus sucking mouse model will help to develop rotavirus vaccine, therapeutic antibody and antiviral drug.
