Purpose: Ketamine is one kind of dissociative anesthetics, in the clinical treatment itcan be used as an anesthetic or surgical anesthetic induction agent, Ketamine has someanalgesic effect of long-term used and it can lead people to appear hallucinogenic effects andaddiction. There are lots of research of Ketamine addiction, but exercise stress which affectketamine addiction of mice have not reported, The contents include:1) by doing theexperiment of conditioned place Preference in mice observed experimentally rewardcharacteristics.2) By adding sports to the procedure of addictive behavior of ketamine on miceto understand the effects of exercise stress on the process of ketamine addiction of mice;3) bydoing behavioral experiments on long-term ketamine feed mice to understand the effect ofketamine on the anxiety and depression behavior of mice.Methods: The conditioned place preference experiments conducted on mice measured thepreliminary CPP value. According to the results of the preliminary tests, the mice weredivided into four groups: control group,1mg/kg ketamine group,3mg/kg ketamine groupand10mg/kg ketamine group then8days of CPP training were done on the mice. On theninth day, the CPP value was measured. Based on the results of the above experiments thegroup of a dose of10mg/kg was chosen to do CPP training and adding motion intervene in thetraining process.Firstly, all mice were divided into four groups: control group (saline), ketamine group (5mg/kg), ketamine group (10mg/kg) and ketamine group (20mg/kg). The four group micewere feed the related drugs accordingly intraperitoneally eight days, feed two times a day, Onthe ninth day, the spontaneous activity, anxious behavior (elevated plus maze test), depressivebehavior (tail suspension test, forced swimming test) were tested.Results:The results of CPP test shows that the CPP value of the ketamine groups (1mg/kg、3 mg/kg、10mg/kg) are all higher than that of the control group;By adding sports to theprocedure of CPP training it is found that the CPP value is lower than that of the ketaminegroup;The results of activity experiment showed that compared with saline group the numberof cavers of small, middle and high dose of ketamine group increases (P>0.05) whichindicates that ketamine can enhance the number of spontaneous activity and the frequency ofcavers;The results of the elevated plus maze experiment found that compared with salinegroup OE value and OT value of the middle and high dose ketamine group is higher than thatof the saline group and there is no difference of OE value and OT value between the salinegroup and the low-dose ketamine group. But; The results of the tail suspension tests showedthat compared with the control group the time period of static of head hanging down of thelow, middle and high dose ketamine group is shorter than that of the saline group;The resultsof the forced swimming test showed that compared with the control group the time period ofstatic of putting head in the water of the low, middle and high dose ketamine group haveextend trend.Conclusion: Ketamine has an increased effect on the trend of spontaneous activity,ketamine played prevent effect on anxiety and depression behavior of mice; ketamine canform drug addiction; movement can inhibit ketamine addiction.
