Turnip is the radix and seed of Brassica rapa L.. The drug is collected in autumn when the radix and seed are ripe. Turnip is also called NIUMA (from Tibetan medicine’s classics Four-Volume Medical Code) in Tibetan herb, and is generally called MANJING, YUANGEN in Tibet. In traditional Tibetan Medicine theory, turnip has the effect of detoxification and nourishing. It is suitable for treatment of all kinds of poisons,’Long’ illness and body weakness. At present, the Tturnip has not established a complete quality control method, turnip polysaccharides have also not been reported effects on mice acute hypoxia.PurposeTo optimize extraction and purification procedures of antioxidant activity polysaccharides from Brassica rapa; Analysis of turnip in the monosaccharide composition of polysaccharide; To study the protective effects of turnip polysaccharide on normobaric hypoxia and acute hypoxic hypoxia injury; To establish a method for quality control of the turnip in order to provide the evidence for health food developing on turnip.Methods1. With the crude polysaccharides yield,the content and antioxidant activity of total polysaccharides as indicators,orthogonal design was used to optimize extraction technology by taking extraction times,solid-liquid ratio and extraction time as factors;With the removal rate of pigment and the loss rate of polysaccharides as comprehensive evaluation index,effects of bleaching temperature,adsorption time and activated carbon dosage on decoloration technology were investigated by orthogonal test.2. Using the high performance liquid chromatography to analyze the monosaccharide composition of polysaccharide.3. Using normobaric hypoxia test and acute hypobaric oxygen chamber hypoxia experiment to study the protective effect of turnip polysaccharide on anoxic injury in mice.4. Using the phenol-sulfuric acid method to determine the total polysaccharide content of turnip.Results1. Optimum extraction technology was as follows:solid-liquid ratio1:30, extracted3times at90℃with2h each time; The best decoloration technology was: absorbed40min with3%activated carbon at60℃. Under these conditions,purity of polysaccharides from B. rapa was up to4.66%, the IC50value of DPPH free radical scavenging was6.71g·L-2. Polysaccharides consisted of mannose, rhamnose. glucuronic acid, galacturonic acid, glucose, galactose, arabonose with the molar ratio of0.26:0.54:0.05:4.17:0.84:1:1.06.3. Turnip polysaccharide high dose group (2.0g·kg-1) could significantly prolong the survival time of hypoxia tolerance of mice,and the effect is quite to0.28g·kg-1positive control group(Rhodiola capsules).4. The turnip high dose group polysaccharide can improve the SOD activity of the mice brain tissue (P<0.05), reduce the generation of MDA mouse brain tissue (P <0.05), increased the SOD activity of serum (P<0.05), reduce the production of MDA in serum (P<0.05); HE staining showed that the turnip polysaccharide high dose group can reduce the mice hypobaric hypoxia damage of brain tissue.5. Based on the characters of turnip, authentication, inspection, content determination and chemical composition analysis, establish a standard Trunip quality standards.6. The total polysaccharide of10batches turnip is in the range of8.76%-11.39%.Conclusions1. These optimized extraction and purification processes were stable and reliable, a high level of polysaccharides with fewer impurities was obtained, which indicated that optimized processes were suitable for industrial production of polysaccharides from B. rapa.Meanwhile,these results indicated that polysaccharides from B. rapa had strong antioxidant activity.and worthy of further study.2. The turnip polysaccharide can significantly protect body injury induced by hypoxia, the role of its significantly improve the body’s free radical scavenging activity, reduce lipid peroxidation injury-related,Prompt turnip polysaccharide has a protective effect on the damage to the body caused by the high altitude.
