Purpose:This research is to explore the pathogenesis of chronic renal failure (CRF) under the guidance of metabolism theory from Huangdi’s Canon of Medicine. It studies and observes "Shensufang"s treatment effect in patients with chronic renal failure early and medium stage through experiment.It studies its treatment mechanism by detecting protein expression from TGF-β1、Smad6、BMP-7、BMP-7R Ⅱ of renal tissue in rats. The research lays foundation for the broader research thinking and effective treatment plan.Methods:Methods of Literature retrieval, literature review and experimental study have been adopted.1. It has reviewed related research about TCM treatment of chronic renal failure for recent10years in CNKI and made induction, analysis and conclusion in the following aspects such as etiology and pathogenesis, syndromedifferentiation,prescription, treatment principle and methods, special recipe for special disease and designed drug treatment.2. Through related literature review of Huangdi’s Canon of Medicine, it has found the core content of "metabolism" theory, and combined with related manifestation of chronic renal failure to explore its basic pathogenesis.3. It has adopted the method of animal experiments. Adenine is used to make model of rat with chronic renal failure. Western medicine, Compounda-KetoAcidTablets and Chinese patent medicine Niaoduqing Keli which are used to cure chronic renal failure are applied to make comparison. Seven groups have been divided including group of Western medicine Compounda-KetoAcidTablets, group of Chinese patent medicine Niaoduqing Keli, group of Chinese medicine Shensufang with high dose, group of Chinese medicine Shensufang with medium dose, group of Chinese medicine Shensufang with low dose, model group and blank group. After treatment, it has observed creatinine and urea nitrogen level in the rat blood, changes of pathological sections in the kidney as well as protein expression of TGF-β Smad6、BMP-7、BMP-7R Ⅱ in the kidney tissue of the rats. Result:This thesis uses "metabolism" theory from Huangdi’s Canon of Medicine as the entry point and studies chronic renal failure from qi transformation of the whole body. In this way, this disease is mainly caused by the gradual decrease or even loss of the qi transformation which leads to the production of pathological products such as water retention and stasis toxin. The water retention and stasis toxin further leads to the disturbance of qi movement which aggravates the imbalance of Zangfu-organs and declining of qi transformation and forms the vicious cycle. In terms of Zangfu-organs, the disease is mainly associated with the liver, the spleen and the kidney. In the early stage, it mainly involves the spleen and in the later stage, the kidney. In terms of the disease nature, it is deficiency in root and excess in manifestation. The animal experiment result:Shensufang can markedly reduce BUN、Scr in the model rats with chronic renal failure. It can improve the pathological changes of renal tubules. It increases protein expression of BMP-7and Smad6in the kidney tissue while decreasing protein expression of TGF-β、which indicate that Shensufang can alleviate renal function damage as well as damage to renal tubule and renal interstitial lesions in model rats.Conclusion:This research discusses the inner meaning of "metabolism" theory from Huangdi’s Canon of Medicine. It studies the theoretical basis of TCM in treating chronic renal failure from the perspective of qi transformation and applies experiment to prove the viability of Shensufang in treating chronic renal failure. Through learning protein expression of renal parenchyma, it explores the mechanism of Shensufang to treat chronic renal failure which provides persuasive theoretical support and experiment basis for Traditional Chinese Medicine to treat chronic renal failure.
