| OBJECTIVE:To explore the effects of jianpiquyu method on VEGF/VEGFR2pathway in retinal pigment epithelium (RPE) of apolipoprotein E-deficient(ApoE-/-) mice by dyslipidemia.METHODS:2-month-old ApoE-/-mice were randomly divided into two big groups:the group of normal chow and the group of high fat chow. And then the former group mice were randomly divided into three groups:the group of2-month-old normal chow(group A);the group of6-month-old normal chow(group B);the control group of7-month-old normal chow (group D). And the latter group mice were randomly divided into three groups:the group of6-month-old high fat chow(group C);the control group of7-month-old high fat chow(group E);the group of7-month-old treatment(group F). Twelve mice in each group.Group A/B/C were into experiment1(model establish),and Group D/E/F were into experiment2(chinese medicine intervention).Group E/F were fed normal chow when the mice6months old,group F mice were gavagely administered with jianpiquyu formula granule for one month.and Group D/E were gavagely administered with same amount of normal saline at the same time. Animal bodymass,blood lipid and hemorheology levels were detected. RPE was observed by Light microscopy.transmission electron microscopy and immunohistochemistry analysed. And the VEGF and VEGFR2were analysed by Western-blot.RESULTS:The animal bodymass,lipid and hemorheology levels in group C were significantly higher than that of group A and group B(P<0.05).The thickness of RPE in group C was statistically lower than that of group A and group B(P<0.05).VEGF and VEGFR2Dyeing area and intergral optical density in group C was higher than that of group A and group B(P<0.05).Protein expression in retina in group C was higher than that of group A and group B(P<0.05). The animal bodymass,lipid and hemorheology levels in group E were significantly higher than that of group D and group F(P<0.05).The thickness of RPE in group D was statistically higher than that of group E and group F(P<0.05), but there is not statistically signigicant between group E and group F (P>0.05).VEGF and VEGFR2Dyeing area and intergral optical density in group E was higher than that of group D and group F(.P<0.05). Protein expression in retina in group E was higher than that of group D and group F(P<0.05).CONCLUSION:1.The dry AMD animal model with aging dyslipidemia was successfully set up by high fat feeding in this study,and it is expected to become AMD experimental research as a more suitable model.2.Jianpiquyu method can slow down cellular damage in RPE, down-regulation the expression of VEGF and VEGFR2level in RPE, regulate and control the VEGF/VEGFR2signal transduction pathways in this animal model.3. It Plays a intervention role in AMD from dry to wet by jianpiquyu method. |
