Objective:To investigate the efficacy of Qingre Mingmu decoction by ultrasonic atomization method in the treatment of acute conjunctivitis.Method:308cases,616eyes of patients diagnosed with acute conjunctivitis based on the patient’s subjective symptoms:foreign body sensation, burning sensation, itching, pain, photophobia, tearing and slit lamp examined under a conjunctival hyperemia, edema, and secretions were randomly dividedinto three groups, by the statistical test, three overall comparable. Treatment group,110cases (220eyes), a control group of108patients with216eyes, the Chinese Medicines90cases (180eyes). The control group with levofloxacin eye drops,4times/day; treatment group on the basis of the control group, inaddition Qingre Mingmu decoction by ultrasonic atomization smoked eye,1day,20min/time; the Chinese Medicines single group Qingre Mingmu decoction by ultrasonic atomization smoked eyes, once a day,20min/time.3day and1week after treatment, the clinical results of patients with symptoms and signs.Results:(1) Treatment group total effective rate was98.19%,the control group t was88.97%, the Chinese Medicines was88.89%;(2) Teatment of the signs and symptoms before and after taking the difference of the average of the comparative analysis, P<0.05,said the difference was significant, the degree of improvement of the overall treatment group to be higher than that of the control group, no significant difference between the control group and TCM group.Conclusion:(1) The acute conjunctivitis treated by ultrasonic atomization method Qingre Mingmu decoction has a clear purpose;(2) Ultrasonic atomization method is scientific and effective.By the ultrasonic atomization method with Qingre Mingmu decoction treatment of acute conjunctivitis in improving symptoms of eye itching, foreign body sensation, burning sensation, and conjunctival irritation effect is obvious;(3) Observation on rabbit conjunctivitis model histopathology, which indicating reducing the inflammatory cells and reducing the conjunctival epithelial cells to stimulate, maintain the goblet cell function, protect the integrity of the ocular surface structure, the prognosis is good.Experimental Research on Conjunctivitis Treated with QingRe MingMu Decoction by Ultrasonic AtomizationMethodPart One Conjunctivitis ModelObjective:To establish an animal model of conjunctivitis can be used to analyze the efficacy and mechanism of action. Method:(1)Establish rabbit conjunctivitis model with micro-injector trickle of cantharidin tincture50ul,1time/d, continuous3d;(2) The end of stimulation after30min, in addition to the normal group, randomly divided into three groups, for the model group, the control group, QingRe MingMu decoction group, n=6.Results:(1)Rabbit eyes after instillation of homemade cantharidin tincture both showed eyes closed, restlessness and other distressing symptoms. Before treatment in addition to the normal group, the animals in each group showed significant inflammatory response,which means modeling success.Conclusion:The method of establishment of rabbit conjunctivitis model is simple and feasible, economical and practical, a short time into a mold, and high molded rate,that can be used to study of QingRe MingMu decoction ultrasonic atomization effect and the mechanism of conjunctivitis modelanalysis.Part Two The Research on Efficacy Studies of Conjunctivitis Model by Ultrasonic Atomization Method with QingRe MingMu DecoctionObjective:To take the study on efficacy of the conjunctivitis model with QingRe MingMu decoction by ultrasonic atomizing method.Methods:(I)Take no eye diseases rabbits after the treatment by ultrasonic nebulizer treatment with Qingre Mingmu decoction.At the limbus at the top of the front puncture room water0.1ml, high performance liquid chromatography in aqueous humor concentration of baicalin,hotlhe cracking/purge and Trap-GC/MS was used to analyze the concentration of the aqueous humor menthol.(2) After modeling success,in addition to the normal group, randomly divided into three groups, for the model group, the control group, QingRe MingMu decoction group, n=6.(3)Last24h after treatment, the animals were sacrificed,to take the left eye of each animal and10%formalin-fixed, paraffin-embedded, sliced, HE staining, and histopathology.Results:(1)The retention time and spectral diagram of the peaks and the baicalin standard color samples in the spectral peaks are inconsistent,that shows no baicalin in the samples.(2)Under the selected chromatographic conditions, chromatographic peak menthol has good separation, and higher retrieval and matching degree and standard atlas, which basically can determine the existence of menthol in samples.(3) Pathological histological observation of QingRe MingMu concentration group, the control group and model group with significant difference (P<005);Conclusions:(1)The use of ultrasonic atomization QingRe MingMu decoction treatment, prescription ingredients menthol by blood-aqueous barrier;(2) Although the aqueous humor concentration test results show that baicalin can not pass the blood-aqueous barrier, the room waterthe larger the baicalin peaks obtained is expected through improved or optimized further study.(3) Observation on rabbit conjunctivitis model histopathology, shows that QingRe MingMu decoction by ultrasonic atomization treated conjunctivitis, reducing the inflammatory cells and reducing the conjunctival epithelial cells to stimulate, at the same time,maintaining the goblet cell function, and protecting the integrity of the ocular surface structure.The prognosis is good.Part Three:The Research of the Mechanism of Treatment of Conjunctivitis Model with QingRe MingMu Decoction by Ultrasonic Atomization MethodObjective:To study analysis of the efficacy of the mechanism of the conjunctivitis model, by ultrasonic atomization with QingRe MingMu decoction.Methods:Rabbits with conjunctivitis were randomly divided into normal group, model group, the control group, QingRe MingMu decoction group, the last treatment24h after the animals were sacrificed,each animal’s left eye with10%formalin-fixed, paraffin-embedded, sliced, immunohistochemical staining observed expression of IL-lbeta, IL-6, TNF-alpha.Results:(1) IL-lbeta control group and TCM concentrations less expression in the epithelial cell layer.(2) IL-6expresses in the conjunctival epithelial cell layer. The control group and Chinese medicine group was compared with the normal group with some difference.(3) TNF-alpha was mainly expressed in the epithelial cell layer.and the collagen fibers also have some expression. Model group on the cortical expression of the most obvious difference compared to the normal group and model group.The control group, the QingRe MingMu decoction group and normal group have little difference.Conclusion:Immunohistochemical localization of rabbit conjunctivitis model of inflammatory factors of IL-1β, IL-6and TNF-α, and qualitative analysis, show that there is a close relationship between conjunctivitis and the factors. The severity of inflammatory factor expression can reflect the conjunctival disease. But if the relationship of conjunctivitis and inflammatory factors were better explained, further research is needed.
