ObjectiveBy the method of flow cytometry, the study aims to illustrate the expressioncharacteristics of the surface molecules CD45RO, CD62L and CCR7on tumorinfiltrating lymphocytes and the distributions of memory T cells subsets in thepatients with HCC and the patients with benign lesions, providing basic theoryfor the study of the memory T lymphocytes’ functions in anti-tumor immunity.MethodsObtained fresh liver tissues from the patients with primary liver cancer andthe patients who underwent hepatectomy because of benign lesions, thenprepared monoplast suspensions and isolated infiltrating lymphocytes fromthem. Used the method of flow cytometry to test the cell surface molecules of Tmemory lymphocytes: CD45RO, CD62L and CCR7..Results1.The proportions of CD4~+T lymphocytes in tissue infiltrating lymphocytes inHCC tissues were (21.38±3.72)%, which were significantly higher than those inadjacent liver tissues (10.12±2.42)%and in normal liver tissues (4.14±4.34)%(P=0.000, P=0.001; P <0.05). The proportions of CD4~+T lymphocytes inadjacent liver tissues were significantly higher than those in normal liver tissuestoo.2. The proportions of CD8~+T lymphocytes in tissue infiltrating lymphocytesin HCC tissues and in adjacent liver tissues were (25.10±5.46)%and(21.70±2.6)%, which were significantly lower than those in normal liver tissues (40.40±13.93)%(P=0.014, P=0.004; P <0.05). The proportions of CD8~+Tlymphocytes were similar between those in HCC tissues and in adjacent livertissues.3. The proportions of CD8~+T lymphocytes in tissue infiltrating lymphocyteswere significantly higher than the proportions of CD4~+T lymphocytes in adjacentliver tissues and in normal liver tissues (t=-7.7, P=0.00<0.05; t=-8.15,P=0.00<0.05), however there were no differences between the proportions ofCD8~+T lymphocytes and CD4~+T lymphocytes in HCC tissues(t=-1.43,P=0.19>0.05).4. The proportions of CD4~+CD45RO~+TCMsubsets in CD4~+CD45RO~+memory T lymphocytes in HCC tissues, adjacent liver tissues and normal livertissues were (0.24±0.22)%,(0.30±0.29)%and (0.73±0.99)%. There were nodifferences between each other’s (P=0.267,0.361,0.913; P>0.05). Theproportions of CD4~+CD45RO~+TEMsubsets in CD4~+CD45RO~+memory Tlymphocytes in HCC tissues, adjacent liver tissues and normal liver tissueswere (88.80±10.79)%,(98.50±0.75)%and (95.27±5.50)%. The proportions ofCD4~+CD45RO~+TEMsubsets in HCC tissues were significantly lower than thosein adjacent liver tissues.5. The proportions of CD8~+CD45RO~+TCMsubsets in CD8~+CD45RO~+memory T lymphocytes in HCC tissues, adjacent liver tissues and normal livertissues were (0.33±0.37)%,(0.27±0.17)%and (0.23±0.31)%. There were nodifferences between each other’s (P=0.553,0.806,0.762; P>0.05). Theproportions of CD8~+CD45RO~+TEMsubsets in CD8~+CD45RO~+memory Tlymphocytes in HCC tissues, adjacent liver tissues and normal liver tissueswere (91.36±8.02)%,(98.54±0.77)%and (98.40±1.78)%. The proportions ofCD8~+CD45RO~+TEMsubsets in HCC tissues were significantly lower than thosein adjacent liver tissues and in normal tissues (P=0.011, P=0.005;P<0.05). Conclusions1. The proportions of CD4~+T lymphocytes in tumor infiltrating lymphocytesof HCC tissues increased evidently, while the proportions of CD8~+T lymphocytesdecreased markedly. The ratios of CD4~+/CD8~+were evidently raised.2. Most of CD45RO~+memory T lymphocytes infiltrating in HCC tissueswere TEM, which played a significant role, while TCMwere less. The proportionsof CD4~+TEMand CD8~+TEMin CD45RO~+memory T lymphocytes in HCC tissuessignificantly lower than non tumor tissues.
