Diphylleia sinensis H. L. Li belongs to Berberidaceae.There is a small bowl piton their roots at each internode and initially this drug was dug in the river high hillsideforest.It was also called “a bowl of water on riverside”.They are mainly distributed inHubei,Shanxi,Gansu,Yunnan,and so on.Its rhizomes and fibrous roots can be used formedicinal.It has function of athermalisation, cooling blood, blood, pain and diarrhea.It often used in the treatment of bruises, rheumatoid arthritis, lumbar and leg pain andirregular menstruation the folk. According to the literature reports,the maincomponent of the root of Diphylleia sinensis H. L. Li has anti-tumor, anti-viral,anti-oxidation, anti-bacterial, anti-inflammatory, anti-radiation as well as pesticidesand other aspects of the pharmacological activity.Objectives: To study the chemical composition of the root of Diphylleia sinensis H. L.Li systematically and especially in antineoplastic physiologically active conductpreliminary studies of the main compound. The isolaed compounds were tested toantitumor activity screening.Methods: The dried root of Diphylleia sinensis H. L. was percolated with95%EtOHand partitioned with ethyl acetate,petroleum ether, n-butyl alcohol fractionally.Constituents were isolated and purified by silica gel column chromatography, TLCpreparation and recrystallization. Their structures were identified on the basis of theirchemical and physical properties as well as the spectra data of MS, IR,UV,1H-NMR,13C-NMR etal. Results: Eighteen compounds had been isolated from the the root of Diphylleiasinensis H.L.Li, they are β-sitosterol(1), podophyllotoxin(2), dehydropodophy-llotoxin(3),4′-demethylpodophyllotoxin(4), isodiphyllin(5),4′-demethyldehy-dropodophyllotoxin(6), picropodophyllin(7), epipodophyllotoxin(8), kaempferol(9),astragalin(10), daucosterol(11), quercetin(12),3,4-dihydroxybenzoic acid(13),deoxypodophyllotoxin(14), picropodophyllotoxin-4-oβ-D-glucoside (15), rutin (16),glucose (17), kaempferol-3-O-β-D-rutinoside (18).Conclusion: Eighteen compounds had been isolated from the the root of DiphylleiasinensisH.L.Li. Compounds1、6、7、8、10、11、16and18were isolated from Diphylleiafor the first time. Compound5was isolated from Berberidaceae for the first time. Inaddition,the compounds from the root of Diphylleia sinensis H. L were conducted apreliminary study of antitumor activity. It provides the evidence for the furtherutilization and research.
