Objective:Construction of standard plasmid of CYP2C19*2mutation detection associated with clopidogrel. Ensure accuracy of genetic test results.Methods:PCR product with genotyping of GG and AA are achieved by specific primers, then they are conjugated with pGM-T vector,the product tested by PCR with plasmid bacterial and then confirmed by DNA sequence,finally the optimized PCR test system are established.Results:The CYP2C19*2product were specifically amplicated by PCR and which were successfully inserted into T vector, they are confirmed by DNA sequence and Sequence Blast.Conclusion:1. Construction of standard plasmid of CYP2C19*2mutation detection associated with clopidogrel.2. Establishment of optimized PCR detection system of CYP2C19.3. It laid the foundation of the internal quality control and external quality assessment of the gene test of CYP2C19.
