Objective: To establish the rats model with alcoholic liver disease, and study the protectiveeffect and mechanism of gypencsides in alcoholic liver disease in rats.Methods:40SD male rats were randomly divided into normal control group10, model groupand drug intervention group15respectively. Model group, alcoholic liver disease model ratswere lavaged with ethanol. Rats in Intervention group were lavaged with gypencsides of200mg/kg at the same time, once a day. After12weeks, all rats were killed and detected the serumtransaminase indicators (ALT, AST), and TNF-α using ELISA kits. Select the part of the livertissue for HE staining and observe the change of its liver histopathology under the opticalmicroscope. Detect the apoptosis of liver homogenate by flow cytometry. Detect TNF alpha byWestwern Blot, detect PPAR alpha, SREBP-1, AMPK by RT(real-time)-PCR.Results: Experimental study, the indicators of model group changed after Gynostemmaintervention., Serum ALT, AST levels of intervention group compared with the model groupdecreased significantly (P <0.05). TNF-α level compared with the model group significantlydecreased (P <0.05). Apoptosis rate of model group was obviously higher than that of normalgroup, the apoptosis rate of intervention group was obviously lower than model group. Theintervention of gypencsides was able to significantly reduced hepatic steatosis and inflammation.Detection sterol regulating element binding protein1(SREBP-1) level significantly decreasedintervention group compared with model group by RT-PCR.Conclusion: gypencsides in rats of alcoholic liver disease liver has better protective effect.Results suggest that gypencsides can significantly reduce liver histological damage and play alarger role in resistance to apoptosis, the mechanism may be realized through lower cytokineTNF-α level. Gynostemma total glucoside has protective effect on alcoholic liver disease modelrats, the results show that the gynostemma total glucoside can significantly reduce serum ALT,AST level, and sterol adjustment element binding protein1(SREBP-1) and TNF alpha level,improve liver function, reduce the liver cells and adipose accumulation. Inhibiting apoptosissignal conduction, give play to the role of anti-inflammatory and anti apoptosis, thus effectivelyprevent the liver damage. Gynostemma total glucoside can reduce the level of TNF alpha andSREBP-1level to prevent effect of alcoholic fatty liver, there is other pathways that TNF alphadirectly acts on the SREBP-1, thus play a role of liver protection.
