Objective:We established the models of proliferative hepatic stellate cell induced by acetaldehyde, then intervened by aqueous extract of Hovenia Dulcis Thunb, to explore the proliferation promotion effect of acetaldehyde on HSC-T6cells, and whether aqueous extract of Hovenia Dulcis Thunb has inhibitory effect on activated cells, by means of observing cell morphology by phase contrast microscope, detecting of cell activity by MTT experiment and identifying the expression of protein a-smooth muscle actin (a-SMA) by Western Blot.Method:Hepatic stellate cell (HSC-T6) was cultured in vitro, then was divided into normal control, model group induced by acetaldehyde (200μ mol/1), and drug intervention group treated with different concentrations of aqueous extract of Hovenia Dulcis Thunb based on model group. The HSC-T6cell induced by acetaldehyde for24hours and treated with different concentrations aqueous extract of Hovenia Dulcis Thunb were observed by phase contrast microscope, cell activity was detected by MTT experiment, and the expression of protein a-smooth muscle actin (a-SMA) was identified by Western Blot, to detect the effect of Hovenia Dulcis Thunb on hepatic stellate cells.Result:1. Observed by phase contrast microscope, the normal control cells grew by static adherence, in the shape of polygon, and had abundant cytoplasm. Model group cells induced by acetaldehyde presented vigorous growth, larger cell body and parapodium stretched out. Drug intervention group treated with different concentrations of aqueous extract of Hovenia Dulcis Thunb based on model group was slow-growing, turned into oval, even exfoliation in high concentration.2. In MTT experiment it turned out that in model group induced by acetaldehyde, cells activity appeared higher than the normal group (P<0.05). Based on model group, when intervened by different concentrations of aqueous extract of Hovenia Dulcis Thunb, HSC-T6cells activity decreased along with concentrations of aqueous extract of Hovenia Dulcis Thunb increase (P<0.05). Treated with some concentrations aqueous extract of Hovenia Dulcis Thunb for different time, cells activity decreased along with time increase (P<0.05)3. Western Blot turned out that in model group induced by acetaldehyde, the expression of protein a-SMA appeared higher than the normal group, and in Drug intervention group, it decreased along with concentrations of aqueous extract of Hovenia Dulcis Thunb increase. Conclusion:1. Acetaldehyde can promote activation and proliferation of hepatic stellate cells. And acetaldehyde, which plays synergy effect with other pro-fibrogenic factors, possibly can aggravate hepatic fibrosis.2. Aqueous extract of Hovenia Dulcis Thunb,which has inhibitory effect on the activation of hepatic stellate cells, maybe play certain therapeutic effect on alcoholic liver fibrosis.
