Expression And Function Of TMEM100in Hepatocellular Carcinoma

Objective:The expression of TMEM100mRNA and protein were detected in hepatocellular carcinoma(HCC) fresh tissue specimens and cell lines. After analysis of differential TMEM100expression in cell lines with different proliferative potential, the relationship of TMEM100expression with development of hepatocellular carcinoma was analyzed.Methods:We randomly selected30pairs of HCC tissue samples from Xiangya Hospital Central-South University. Normal liver cell line and four liver cancer cell lines with different proliferative capacity were cultured. Real-time PCR, semi-quantitative RT-PCR and Western-blotting were performed to detect the expression of TMEM100in the HCC tissues and cell lines.Results:1. HCC tissues experiments:Real-time PCR results showed that TMEM100were low expression in90%HCC tissues compared to ANLTs. In addition, the result of semi-quantitative RT-PCR and Western-blotting were consistent with the Real-time PCR results.2.cell experiments:Real-time PCR results showed TMEM100mRNA expression was significantly higher than that in the normal liver cell line LO2. Furthermore, the expression of TMEM100in SMMC-7721and HepG2which have high proliferative capacity were significantly lower than those cell lines with low proliferation capacity like HCCLM3and MHCC97-L. Also, the results of semi-quantitative RT-PCR and Western-blotting were consistent with Real-time PCR.3.Prognosis:The survival time of patients with high TMEM100expression was longer than those with low TMEM100expression.Conclusion:1. TMEM100expression was significantly lower in HCC, which suggesting it may be related to the tumorigenesis and development of HCC. And its low expression may indicate poor prognosis for HCC patients.2.Over expression of TMEM100probably suppressed proliferation of hepatocellular carcinoma.
