Association Of Toll-like Receptor9Genetic Polymorphism With Spontaneous Viral Clearance Of Hepatitis B Virus Infection And Reactivity Of Liver Disease

Objective:To discuss the correlation between the single nucleotide polymorphisms(SNPs) of Toll-like receptor9(TLR9) gene and clinical status of Hepatitis B virus infection.Methods:Collect422patients with persisitent chronic HBV infection which included149asymptomatic carriers(ASC),178chronic hepatitis B(CHB) patients and95patients associated acute on chronic liver failure(ACLF), and174cases of HBV spontaneous clearance persons. The four TLR9gene SNPs at positions rs187084, rs352139, rs352140and rs5743836were analyzed by the Taqman probe and real-time polymerase chain reaction and the genotypic distribution was analyzed according disease progression of HBV infeciton, inflammation degree and HBV-DNA loads.Results:TLR9rs187084、rs352139、rs352140genotyper and allele distribution were different between ASC group and contol(P<0.05). The frequency of rs187084genotype TT、TC and allele T of HBV spontaneous clearance persons were significantly higher than those of chronic HBV infection (P<0.05). In the chronic HBV infection group, The frequency of rs187084genotype TT、TC and allele T of CHB and ACLF group were significantly higher than those of ASC group (P<0.05), genotype AA、allele A of rs352139and genotype CC、allel C of rs352140of CHB group were obviously higher than those of ASC group. The three SNPs analysis of rs187084、rs352139、rs352140have no obvious correlation with the inflammation degree of liver and serum HBV-DNA loads. The study also found that rs5743836may not exist gene polymorphism in this crowd of this study. And there was a relationship of Linkage disequilibrium between rs187084、rs352139and rs352140.Conclusion:1、TLR9gene rs187084、rs352139、rs352140polymorphism is associated with the body’s immune clearance of HBV infection and liver inflammation activity, but has no obvious correlation with inflammation degree of liver and serum HBV-DNA loads.2、 rs5743836may not exist gene polymorphism in this crowd of this study.3、There was a relationship of Linkage disequilibrium among rs187084、rs352139and rs352140.
